Western Blot: Vimentin Antibody [NB300-223] - Analysis of tissue and cell lysates. Antibody at 1:5000 in red. [1] protein standard (red), [2] rat whole brain lysate, [3] HeLa, [4] SH-SY5Y, [5] HEK293, and [6] NIH-3T3 cell lysates. NB300-223 binds to the vimentin protein showing a single band at ~50 kDa. The blot was simultaneously probed with mouse mAb to MAP2C/D, dilution 1:5000 in green, revealing multiple bands around 280 kDa that correspond to full length MAP2A/2B isotypes, and ~70 kDa bands which are MAP2C/D isotypes. MAP2 isotypes are seen only in extracts containing neuronal lineage cells.
Immunocytochemistry/Immunofluorescence: Vimentin Antibody [NB300-223] - Human A7 cells. Cells were fixed with 10% formalin and permeabilized with 0.1% TritonX-100. Immunohistochemistry-Paraffin: Chromogranin A Antibody [NB120-15160] - IHC analysis of a formalin fixed paraffin embedded tissue section of mouse pancreas using Chromogranin A antibody at a dilution of 1:500. The signal was developed using HRP-conjugated anti-rabbit secondary and DAB reagent which followed counterstaining of the section with hematoxylin. The antibody generated a strong staining in Islets of Langerhans. Also, in the periphery of the cells, the pattern of staining was punctate representing the vesicles with Chromogranin A. Cells of lobules and acini showed a weak cytoplasmic and nuclear staining which is potentially the secreted pool of Chromogranin A.

The cells were incubated with anti-Chromogranin A Antibody at 2 ug/ml overnight at 4C and detected with an anti-rabbit Dylight 488 (Green) at a 1:500 dilution. Nuclei were counterstained with DAPI (Blue). Cells were imaged using a 40X objective.

Western Blot: S100B Antibody (15F4NB) [NBP2-45224] - Total protein from human brain (lane 1) and mouse brain (lane 2) was separated on a 4-15% gel by SDS-PAGE, transferred to 0.2 um PVDF membrane for 30 minutes and blocked in 5% non-fat milk in TBST. The membrane was probed with 2 ug/ml anti-S100B in 1% milk and detected with an anti-mouse HRP secondary antibody using chemiluminescence.
Immunocytochemistry/Immunofluorescence: S100B Antibody (15F4NB) [NBP2-45224] - A431 cells were fixed for 10 minutes using 10% formalin and then permeabilized for 5 minutes using 1X TBS + 0.5% Triton-X100. The cells were incubated with S100B (15F9NB) at a 1:100 dilution overnight at 4 degrees Celsius and detected with Dylight 488 (Green) at a 1:500 dilution. Actin was detected with Phalloidin 568 (Red) at a 1:200 dilution. Nuclei were detected with DAPI (Blue) at 2.0 ug/ml in 1X PBS. Cells were imaged using a 40X objective. Image using the Azide Free form of this antibody.

PVDF membrane was probed with 1 µg/mL of Sheep Anti-Human CEACAM‑7 Antigen Affinity-purified Polyclonal Antibody (Catalog # AF4478) followed by HRP-conjugated Anti-Sheep IgG Secondary Antibody (Catalog # HAF016). A specific band was detected for CEACAM‑7 at approximately 45 kDa (as indicated). This experiment was conducted under reducing conditions and using Immunoblot Buffer Group 8.
CEACAM‑7 was detected in immersion fixed paraffin-embedded sections of human colon cancer tissue using Sheep Anti-Human CEACAM‑7 Antigen Affinity-purified Polyclonal Antibody (Catalog # AF4478) at 3 µg/mL overnight at 4 °C. Tissue was stained using the Anti-Sheep HRP-DAB Cell & Tissue Staining Kit (brown; Catalog # CTS019) and counterstained with hematoxylin (blue). Immunohistochemistry-Paraffin: MUC-1 Antibody (SM3) [NB120-22711] - IHC analysis of formalin fixed paraffin embedded tissue section of human breast cancer xenograft using MUC-1 antibody clone SM3 at 1:10 dilution. The xenograft section depicted a very specific and intense signal in the periphery of the cancer cells. The necrotic cells also developed a strong immune-positivity while the tumor stroma as well as the nuclei of cells were negative for immunostaining.
