• Specificity: This monoclonal antibody recognizes extracellular epitope 2 within the N-terminal Ig domain of human CD147. It is expressed more intensely on thymocytes than on mature peripheral blood T cells. CD147 is important in spermatogenesis, embryo implantation, neural network formation, and tumor progression. It stimulates the production of interstitial collagenase, gelatinase A, stromelysin-1 and various metalloproteinases (MMPs) by fibroblasts. These enzymes are important factors in cancer invasion and metastasis.

• Application Notes: Immunohistochemistry (Formalin-fixed): 1-2ug/ml for 30 minutes at RT. Staining of formalin-fixed tissues requires heating tissue sections in 10mM Tris with 1mM EDTA, pH 9.0, for 45 min at 95C followed by cooling at RT for 20 minutes.
  Inhibits Homotypic Aggregation, Adhesion to Matrix Proteins, and Migration through Matrigel: Order Ab without BSA/Azide.
  Optimal dilution for a specific application should be determined.

Background

CD147 plays a pivotal role in spermatogenesis, embryo implantation, neural network formation and tumor progression. It stimulates adjacent fibroblasts to produce matrix metalloproteinases (MMPS). CD147 may target monocarboxylate transporters SLC16A1, SLC16A3 and SLC16A8 to plasma membranes of retinal pigment epithelium and neural retina. CD147 also seems to be a receptor for oligomannosidic glycans. In vitro, it promotes outgrowth of astrocytic processes. CD147 forms homooligomers in a cis-dependent manner on the plasma membrane. It forms a complex with MMP1 at the tumor cell surface. CD147 interacts with SLC16A1 and SLC1A3; probably a BSG dimer is associated with a monocarboxylate transporter dimer. CD147 also interacts with ATP1B2, MAG and L1CAM.
