Background

Dickkopf related protein 1 (Dkk-1), along with Dkk -2, -3, -4, and Soggy, are secreted proteins with two conserved cysteine-rich domains. Mature human Dkk-1 is a 40 kDa glycosylated protein that shares 86% aa sequence identity with mouse and rat Dkk-1. Dkk-1 antagonizes Wnt signaling by forming ternary complexes with LRP5/6 and Kremen1 or Kremen2. Its function is important during the development of limbs, neurons, hair follicles, retina, and bone.
