Specificity
Cytokeratin 8 (CK8) belongs to the type II (or B or basic) subfamily of high molecular weight cytokeratins and exists in combination with cytokeratin 18 (CK18). This MAb cocktail recognizes all simple epithelia including glandular epithelium, for example thyroid, female breast, gastrointestinal tract, respiratory tract, and urogenital tract including transitional epithelium. All adenocarcinomas and most squamous carcinomas are positive but keratinizing squamous carcinomas are usually negative. This antibody is useful in demonstrating the presence of Paget cells; there is very little keratin 18 in the normal epidermis so only Paget cells are stained.Immuno-histochemical staining with this MAb is indistinguishable from that obtained with monoclonal antibody 5D3.

Immunohistochemistry-Paraffin: Cytokeratin 1 Antibody (LHK1) [NB100-2756] - IHC analysis of a formalin fixed paraffin-embedded (FFPE) human breast cancer using 5ug/ml conc. of Cytokeratin 1 antibody (clone LHK1) on a Bond Rx autostainer (Leica Biosystems). The assay involved 20 minutes of heat induced antigen retrieval (HIER) using 10mM sodium citrate buffer (pH 6.0) and endogenous peroxidase quenching with peroxide block. The sections were incubated with primary antibody for 30 minutes and Bond Polymer Refine Detection (Leica Biosystems) with DAB was used for signal development followed by counterstaining with hematoxylin. Whole slide scanning and capturing of representative images (20X) was performed using Aperio AT2 (Leica Biosystems). A membrane-cytoplasmic staining of Cytokeratin 1 was observed in the cancer cells with signal localized more to the cell membranes. Tumor cores showed a decreased staining and tumor stroma was mostly negative for Cytokeratin 1.

Immunohistochemistry-Paraffin: MUC-1 Antibody (SM3) [NB120-22711] - IHC analysis of formalin fixed paraffin embedded tissue section of human breast cancer xenograft using MUC-1 antibody clone SM3 at 1:10 dilution. The xenograft section depicted a very specific and intense signal in the periphery of the cancer cells. The necrotic cells also developed a strong immune-positivity while the tumor stroma as well as the nuclei of cells were negative for immunostaining.

Immunohistochemistry-Paraffin: p53 Antibody (PAb 240) [NB200-103] - p53 was detected in immersion fixed paraffin-embedded sections of human prostate cancer using anti-human mouse monoclonal antibody (Catalog # NB200-103) at 1:200 dilution overnight at 4C. Tissue was stained using the VisuCyte anti-mouse HRP polymer detection reagent (Catalog # VC001) with DAB chromogen (brown) and counterstained with hematoxylin (blue). Western Blot: Vimentin Antibody [NB300-223] - Analysis of tissue and cell lysates. Antibody at 1:5000 in red. [1] protein standard (red), [2] rat whole brain lysate, [3] HeLa, [4] SH-SY5Y, [5] HEK293, and [6] NIH-3T3 cell lysates. NB300-223 binds to the vimentin protein showing a single band at ~50 kDa. The blot was simultaneously probed with mouse mAb to MAP2C/D, dilution 1:5000 in green, revealing multiple bands around 280 kDa that correspond to full length MAP2A/2B isotypes, and ~70 kDa bands which are MAP2C/D isotypes. MAP2 isotypes are seen only in extracts containing neuronal lineage cells.

Simple Western lane view shows lysates of HepG2 human hepatocellular carcinoma cell line, loaded at 0.2 mg/mL. A specific band was detected for alpha ‑Fetoprotein/AFP at approximately 70 kDa (as indicated) using 5 µg/mL of Mouse Anti-Human/Mouse alpha ‑Fetoprotein/AFP Monoclonal Antibody (Catalog # MAB1368). This experiment was conducted under reducing conditions and using the12-230 kDa separation system.
