Western Blot: Vimentin Antibody [NB300-223] - Analysis of tissue and cell lysates. Antibody at 1:5000 in red. [1] protein standard (red), [2] rat whole brain lysate, [3] HeLa, [4] SH-SY5Y, [5] HEK293, and [6] NIH-3T3 cell lysates. NB300-223 binds to the vimentin protein showing a single band at ~50 kDa. The blot was simultaneously probed with mouse mAb to MAP2C/D, dilution 1:5000 in green, revealing multiple bands around 280 kDa that correspond to full length MAP2A/2B isotypes, and ~70 kDa bands which are MAP2C/D isotypes. MAP2 isotypes are seen only in extracts containing neuronal lineage cells.

This experiment was conducted under reducing conditions and using the12-230 kDa separation system.
Western blot shows lysates of HepG2 human hepatocellular carcinoma parental cell line and alpha &amp;#8209;Fetoprotein/AFP knockout HepG2 cell line (KO). PVDF membrane was probed with&amp;nbsp;0.1&amp;nbsp;µg/mL of Mouse Anti-Human/Mouse alpha ‑Fetoprotein/AFP Monoclonal Antibody (Catalog # MAB1368) followed by HRP-conjugated Anti-Mouse IgG Secondary Antibody (Catalog&amp;nbsp;# HAF018). A specific band was detected for alpha ‑Fetoprotein/AFP at approximately 70&amp;nbsp;kDa (as indicated) in the parental HeLacell line, but is not detectable in knockout HeLa cell line. Whole cell protein from Jurkat cells treated with and without 2 uM staurosporine as indicated was separated on a 4-15% gel by SDS-PAGE, transferred to 0.2 um PVDF membrane and blocked in 5% non-fat milk in TBST. The membrane was probed with 5 ug/ml anti-Caspase 3 in 1% milk, and detected with an anti-mouse HRP secondary antibody using a Femto sensitivity chemiluminescence reagent. Note the detection of both pro-caspase 3 at 35 kDa and the cleaved active caspase 3 at 15-17 kDa.
Immunohistochemistry-Paraffin: Caspase-3 Antibody (31A1067) - (Pro and Active) [NB100-56708] - Tissue section of human spleen using 1:200 dilution of Caspase-3 antibody (clone 31A1067). The staining was developed with HRP labeled anti-mouse IgG secondary antibody and DAB reagent, and nuclei of cells were counter-stained with hematoxylin. This Caspase 3 antibody generated primarily a specific cytoplasmic staining in a subset of spleenocytes with some nuclear signal in a few cells.
