Western Blot: Ctip1 Antibody [NB600-258] - Detection of Human Ctip1 (Bcl11a) by western blot. Samples: Whole cell lysate (5, 15 and 50 ug) from Ramos cells. Antibodies: Affinity purified rabbit Ctip1 Antibody NB600-258 used at 0.4 ug/ml. Detection: Chemiluminescence with exposure times of 3 minutes.

Immunoprecipitation: Ctip1 Antibody [NB600-258] - Detection of human Ctip1 by western blot of immunoprecipitates. Samples: Whole cell lysate (1.0 mg per IP reaction; 20% of IP loaded) from Ramos cells. Antibodies: Affinity purified rabbit Ctip1 Antibody NB600-258 used for IP at 6 ul/mg lysate. Ctip1 was also immunoprecipitated by rabbit anti-Ctip1 antibodies (NB600-259, NB600-261) and rabbit anti-Ctip1 recombinant monoclonal antibody [BLR073G] (NBP2-78710). For blotting immunoprecipitated Ctip1, NBP2-78710 was used at 1:1000. Chemiluminescence with an exposure time of 75 seconds.

Background

The B cell leukemia 11A protein (BCL11A/Evi9/CTIP1) has been implicated in hematopoietic cell development and malignancies. BCL11A is a transcriptional repressor that binds directly to a GC-rich motif and is also recruited to a promoter template via interaction with the orphan nuclear receptor, chicken ovalbumin upstream promoter transcription factor II. In both cases, BCL11A-mediated transcriptional repression is only minimally reversed by trichostatin A, suggesting the possible lack of involvement of class I or II histone deacetylases. Nonetheless, chromatin immunoprecipitation assays revealed that expression of BCL11A in mammalian cells resulted in deacetylation of histones H3 and/or H4 that were associated with the promoter region of a reporter gene. BCL11A mediated transcriptional repression, as well as deacetylation of histone H3/H4 in BCL11A-transfected cells, was partially reversed by nicotinamide, an inhibitor of class III histone deacetylases such as SIRT1. SIRT1 was found to interact directly with BCL11A and was recruited to the promoter template in a BCL11A-dependent manner leading to transcriptional repression. These findings indicate a role for SIRT1 in transcriptional repression mediated by BCL11A in mammalian cells.

1. Does Ctip1 Ab (NB600-258) recognizes all isoforms of the BCL11a (S,L,XL)? why is the Band size showing 117 kDa instead of 91 kDa?
   • NB600-258 was raised against the first 50 residues of Ctip1 which based on uniron is a conserved and unaltered region in all isoforms of the protein. However, we have not tested nor confirmed that this antibody recognizes any isoforms other than the canonical sequence. Also, a differential size of the protein on Western blot from what is calculated can often be attributed to post-translational modifications.
