Complement Component C5aR1 Antibody (32-G1) [CoraFluor™ 1]

CoraFluor™ 1, amine reactive (Catalog:7920) and CoraFluor™ 2, amine reactive (Catalog # 7950) are terbium-based probes that have been developed for use as TR-FRET donors. They emit wavelengths compatible with commonly used fluorescent acceptor dyes such as BODIPY® (or BDY) and Janelia Fluor® dyes, FITC (Catalog # 5440), TMR and Cyanine 5 (Catalog # 5436). CoraFluor™ fluorescence is brighter and more stable in biological media than existing TR-FRET donors, leading to enhanced sensitivity and improved data generation. CoraFluor™ 1 exhibits excitation upon exposure to a 337 nm UV laser.

Complement Component C5aR1 Antibody (32-G1) [CoraFluor™ 1] Summary

• Description: CoraFluor(TM) 1 is a high performance terbium-based TR-FRET (Time-Resolved Fluorescence Resonance Energy Transfer) or TRF (Time-Resolved Fluorescence) donor for high throughput assay development. CoraFluor(TM) 1 absorbs UV light at approximately 340 nm, and emits at approximately 490 nm, 545 nm, 585 nm and 620 nm. It is compatible with common acceptor dyes that absorb at the emission wavelengths of CoraFluor(TM) 1. CoraFluor(TM) 1 can be used for the development of robust and scalable TR-FRET binding assays such as target engagement, ternary complex, protein-protein interaction and protein quantification assays.

• Immunogen: This antibody was raised by immunizing mice a synthetic Complement Component C5aR1 C-terminal phosphopeptide that contained phosphoserine instead of serine at position 334 and also at the neighboring phosphorylation sites Ser-332 and Ser-338.
• Specificity: The 32-G1 antibody recognizes Complement Component C5aR1 which has its serine 334 phosphorylated. The mAb 32-G1 reacted specifically with a Complement Component C5aR1 peptide that contained phosphoserine 334 and also reacted with Complement Component C5aR1s that were isolated from C5a-stimulated, transfected RBL-Complement Component C5aR1 cells.

Background

The human anaphylatoxin C5a is a 74 residue glycopeptide which is generated by proteolytic cleavage of the complement factor C5 in the course of complement activation. A variety of biological effects evoked by C5a can be demonstrated, rendering this molecule an important mediartor of inflammation, with granulocytes and macrophages as the main target cells. All cellular responses to C5a are specifically mediated by interactions with the membrane bound C5a receptor, a seven transmembrane GTP binding coupled receptor that belongs to the rhodopsin supergene family. C5a receptor 1 expression has been reported in myeloid blood cells, brain, liver, lung, spleen, heart, kidney, and intestinal tract. ESTs have been isolated from B cell/lung/testis, nerve, and placenta libraries.
