Western Blot: CNPase Antibody (1H10) [NBP2-50031] - Western blot analysis of different tissue lysates using mouse mAb to CNP, NBP2-50031, dilution 1:2,000 in green: [1] protein standard (red), [2] rat brain, [3] rat spinal cord, [4] mouse brain, [5] mouse spinal cord. A doublet of bands at 46 and 48kDa correspond to the major isotypes of the CNP protein.

Immunocytochemistry/Immunofluorescence: CNPase Antibody (1H10) [NBP2-50031] - Mixed neuron-glial cell cultures stained with NBP2-50031 (red) and polyclonal antibody against GFAP (green, NB300-141). NBP2-50031 stains strongly in oligodendrocytes, whereas GFAP labels only the intermediate filaments in astrocytes. Blue is DNA staining.

Immunohistochemistry Free-Floating: CNPase Antibody (1H10) [NBP2-50031] - Analysis of rat cerebellum section stained with mouse mAb to CNP, NBP2-50031, dilution 1:500 in green and costained with chicken pAb to NF-M, dilution 1:1,000, in red. The blue is DAPI staining of nuclear DNA. Following transcardial perfusion of rat with 4% paraformaldehyde, brain was post fixed for 24 hours, cut to 45uM, and free-floating sections were stained with above antibodies. The CNP antibody specifically stains oligodendrocytes, cells that create the myelin sheath around axons. NF-M antibody labels axons of neuronal cells.

Background

2,3-cyclic nucleotide-3-phosphodiesterase (CNP) is a membrane bound, microtubule associated protein that is among the most abundant myelin proteins of the CNS. It is thought that CNP may serve as a regulator of tubulin polymerization and of microtubule distribution (Bifulco et al., 2002). It was recently found that CNP may also function as a possible linker protein anchoring microtubules to the plasma membrane via a 13 residue C-terminal CNP fragment (Bifulco et al., 2002, Esposito et al., 2008).
