  1. Assay Diluent A (Item D): Diluent buffer, 0.09% sodium azide as preservative.
  2. Assay Diluent B (Item E): 5x concentrated buffer.
  3. Detection Antibody TECK (Item F): Biotinylated anti-Mouse TECK. Each vial is enough to assay half the microplate.
  4. HRP-Streptavidin Concentrate (Item G): 120x concentrated HRP-conjugatedstreptavidin.
  5. Standard Protein (Item C): Mouse TECK. 1 vial is enough to run each standard in duplicate.
  6. Stop Solution (Item I): 0.2 M sulfuric acid.
  7. TECK Microplate (Item A): Coated with anti-Mouse TECK.
  8. TMB One-Step Substrate Reagent (Item H): 3, 3’, 5, 5’- tetramethylbenzidine (TMB) in buffered solution.
  9. Wash Buffer Concentrate (20X) (Item B): 20X concentrated solution.

Background

TECK belongs to the subfamily of small cytokine CC genes. Cytokines are a family of secreted proteins involved in immunoregulatory and inflammatory processes. The CC cytokines are proteins characterized by two adjacent cysteines. The cytokine encoded by this gene displays chemotactic activity for dendritic cells, thymocytes, and activated macrophages but is inactive on peripheral blood lymphocytes and neutrophils. The product of this gene binds to chemokine receptor CCR9.
