  1. Assay Diluent A (Item D): Diluent buffer, 0.09% sodium azide aspreservative.
  2. Assay Diluent B (Item E): 5x concentrated buffer.
  3. Detection Antibody MPIF-1 (Item F): Biotinylated anti-human MPIF-1 (eachvial is enough to assay half microplate).
  4. HRP-Streptavidin Concentrate (Item G): 200x concentrated HRP-conjugatedstreptavidin.
  5. MPIF-1 Microplate (Item A): 96 wells (12 strips x 8 wells) coated withanti-human MPIF-1.
  6. Standard Protein (Item C): Human MPIF-1. 1 vial is enough to run eachstandard in duplicate.
  7. Stop Solution (Item I): 0.2 M sulfuric acid.
  8. TMB One-Step Substrate Reagent (Item H): 3,3’,5,5’-tetramethylbenzidine(TMB) in buffer solution.
  9. Wash Buffer Concentrate (20X) (Item B): 20X concentrated solution.

Background

Macrophage Inflammatory Protein 3, also known as CCL23, is located on the p-arm of chromosome 9 with other CC cytokine genes which are involved in immunoregulatory and inflammatory processes. In patients with rheumatoid arthritis, high levels on CCL23(19-99) have been found in synovial fluids. This protein is known to have interaction with CCR1, CCR10, CCR2, CCR3 and CCR4.
