HRP conjugated goat anti-rabbit IgG (H+L) cross adsorbed secondary antibody was used with ECL substrate for the detection of Bcl2 antibody bound to the blotted protein. This Bcl2 antibody detected a specific band at expected molecular weight marker position of Bcl2 protein (26kDa).

PVDF membrane was probed with 1 µg/mL of Mouse Anti-Human Phospho-FADD (S194) Monoclonal Antibody (Catalog # MAB7047) followed by HRP-conjugated Anti-Mouse IgG Secondary Antibody (Catalog # HAF007). A specific band was detected for FADD at approximately 28 kDa (as indicated). This experiment was conducted under reducing conditions and using Immunoblot Buffer Group 1.

Cells were stained using the NorthernLights™ 557-conjugated Anti-Mouse IgG Secondary Antibody (red; Catalog # NL007) and counterstained with DAPI (blue). Specific staining was localized to nuclei. The cells were incubated with anti-TRAIL R2 (H76) at 5ug/ml overnight at 4C and detected with an anti-rabbit Dylight 488 (Green) at a 1:500 dilution. Alpha tubulin (DM1A) NB100-690 was used as a co-stain at a 1:1000 dilution and detected with an anti-mouse Dylight 550 (Red) at a 1:500 dilution. Nuclei were counterstained with DAPI (Blue). Cells were imaged using a 40X objective.

PVDF membrane was probed with 1 µg/mL of Goat Anti-Human TRAIL R1/TNFRSF10A Antigen Affinity-purified Polyclonal Antibody (Catalog # AF347) followed by HRP-conjugated Anti-Goat IgG Secondary Antibody (Catalog # HAF019). A specific band was detected for TRAIL R1/TNFRSF10A at approximately 50 kDa (as indicated). This experiment was conducted under reducing conditions.
In the presence of a cross-linking antibody, Mouse polyHistidine Monoclonal Antibody (Catalog # MAB050) and the metabolic inhibitor actinomycin D (1 µg/mL), Recombinant Human TRAIL R1/TNFRSF10A Fc Chimera (Catalog # 347‑DR) inhibits Recombinant Human TRAIL/TNFSF10 (Catalog # 375‑TL) induced cytotoxicity in the L‑929 mouse fibroblast cell line in a dose-dependent manner (orange line), as measured by crystal violet staining. Under these conditions, inhibition of Recombinant Human TRAIL/TNFSF10 (12 ng/mL) activity elicited by Recombinant Human TRAIL R1/TNFRSF10A Fc Chimera (10 ng/mL) is neutralized (green line) by increasing concentrations of Goat Anti-Human TRAIL R1/TNFRSF10A Antigen Affinity-purified Polyclonal Antibody (Catalog # AF347). The ND50 is typically 0.02‑0.055 µg/mL.
TRAIL R1/TNFRSF10A was detected in immersion fixed paraffin-embedded sections of human brain using Goat Anti-Human TRAIL R1/TNFRSF10A Antigen Affinity-purified Polyclonal Antibody (Catalog # AF347) at 10 µg/mL overnight at 4 °C. Tissue was stained using the Anti-Goat HRP-DAB Cell & Tissue Staining Kit (brown; Catalog # CTS008) and counterstained with hematoxylin (blue).   1. May we ask the MW difference between the active form and the proform of Caspase 8 while performing WB?
    • In treated cells induced to undergo apoptosis, caspase-8 migrates as a 55/53 kDa (pro-form), 41/42 kDa (a cleaved/active or intermediate form), and 18 kDa (active form). The proform (55/53 kDa) is still seen in treated cells because not all cells undergo apoptosis at once.
