Immunoprecipitation: CARKL Antibody [NBP2-61223] - Detection of Human SHPK by Western Blot of Immunoprecipitates. Samples: Whole cell lysate (1.0 mg per IP reaction; 20% of IP loaded) from 293T cells prepared using NETN lysis buffer. Antibodies: Affinity purified rabbit anti-SHPK antibody NBP2-61223 used for IP at 8 ul per reaction. SHPK was also immunoprecipitated by rabbit anti-SHPK antibody. For blotting immunoprecipitated SHPK, NBP2-61223 was used at 1:400. Detection: Chemiluminescence with an exposure time of 3 minutes.

Background

CARKL is encoded by this gene has weak homology to several carbohydrate kinases, a class of proteins involved in the phosphorylation of sugars as they enter a cell, inhibiting return across the cell membrane. Sequence variation between this novel gene and known carbohydrate kinases suggests the possibility of a different substrate, cofactor or changes in kinetic properties distinguishing it from other carbohydrate kinases. The gene resides in a region commonly deleted in cystinosis patients, suggesting a role as a modifier for the cystinosis phenotype. The genomic region is also rich in Alu repetitive sequences, frequently involved in chromosomal rearrangements. [provided by RefSeq]
