Bone morphogenic protein 4 (BMP4) is a member of the TGF-beta superfamily that plays a critical role in mammalian mesenchymal development, influencing hematopoeisis and osteoblast differentiation and function. BMP-4 is secreted as 54 kDa inactive precursor peptides that dimerize and are activated following cleavage by members of the proprotein convertase family. Active BMP-4 binds type I and type II serine/threonine kinase receptors which signal via phosphorylation of Smad family proteins. BMP-4 and the highly homologous BMP-2 are functional orthologues of the decapentaplegic (dpp) gene in Drosophila that regulates embryonic leg and wing imaginal disc patterning and larval gut development.
