Dot: concentration determined using recombinant NGFB at 400 ng under non- and 200 ng under reducing conditions. For capture ELISA, Monoclonal Anti-Human Nerve Growth Factor-Beta can be used as the capture antibody. Determined in combination with a biotinylated NGF-Beta polyclonal detection antibody at 50-100 ng/ml (100 ul/well), an ELISA for sample volumes of 100 ul with a range of 31.2 to 2000 pg/ml may be obtained. For capture ELISA, a working concentration of 1-2 ug/ml is determined to coat plates at 100 ul/well. Inhib: Use at a concentration of 1 - 2 ug/ml. The Neutralization Dose50 (ND50) for anti-human NGFB is approximately 1-3 ug/ml in the presence of 5 ng/ml of recombinant human NGFB using the NGFB responsive cell line TF-1.
This clone has been shown to neutralize the biological activity of recombinant human NGFB and recombinant rat NGFB, but not recombinant mouse NGFB. When used as a capture antibody in ELISAs, this antibody shows less than 5% cross-reactivity with recombinant rat NGFB and mouse NGFB.Not yet tested in other species. Please note that this antibody is reactive to Mouse and derived from the same host, Mouse. Additional Mouse on Mouse blocking steps may be required for IHC and ICC experiments. Background
Nerve growth factor (NGF) is a polypeptide involved in the regulation of growth and differentiation of sympathetic and some sensory neurons, and is composed by 3 types of subunits (alpha, beta and gamma) that specifically interact to form a 7S, 130 kDa molecular weight complex. The beta-NGF subunit is a secreted protein that forms a homodimer before being incorporated into the NGF complex. The human and mouse sequences are very similar and mutations in this gene have been associated with hereditary sensory and autonomic neuropathy, type 5 (HSAN5). Deregulation of its expression has been associated with allergic rhinitis.
