Flow (Intracellular): SR-BI Antibody [NB400-101] - An intracellular stain was performed on HeLa cells with NB400-101AF647 (blue) and a matched isotype control (orange). Cells were fixed with 4% PFA and then permeablized with 0.1% saponin. Cells were incubated in an antibody dilution of 2.5 ug/mL for 30 minutes at room temperature. Both antibodies were conjugated to Alexa Fluor 647.

Immunocytochemistry/Immunofluorescence: SR-BI Antibody [NB400-101] - HeLa cells were fixed and permeabilized for 10 minutes using -20C MeOH. The cells were incubated with anti-SR-BI at 2 ug/ml overnight at 4C and detected with an anti-rabbit DyLight 488 (Green) at a 1:500 dilution. Alpha tubulin (DM1A) NB100-690 was used as a co-stain at a 1:1000 dilution and detected with an anti-mouse DyLight 550 (Red) at a 1:500 dilution. Nuclei were counterstained with DAPI (Blue). Cells were imaged using a 40X objective.

Western Blot: IL-1 beta/IL-1F2 Antibody [NB600-633] - Antibody was used at a 1:200 dilution incubated 1h at room temperature to detect dog IL-1b by Western blot. Lane 1 and 2 were loaded with 2.5 ug and 500 ng of dog IL-1b respectively. The molecular weight of the detected band is estimated by comparison to molecular weight markers (not shown). Detection occurred using a 1:3,000 dilution of IRDye (TM) 800 conjugated Donkey anti-Rabbit IgG for 1h at room temperature.

Effects of butyrate treatment on curdlan-induced SpA mice. SKG mice were injected with 3 mg of curdlan through intraperitoneal and oral administration. One week later, sodium butyrate (100 mg/kg) was administered orally once a day for 14 weeks. (A) Arthritis score and incidence were monitored by observation twice a week. Tissue section of human intestine using Caspase-1 antibody (clone 14F468) at 5ug/ml concentration (1:200 dilution). The primary antibody binding to Caspase 1 in cells was detected using HRP conjugated anti-Mouse secondary antibody with DAB reagent, and the sections were further counterstained with hematoxylin for labeling cellular nuclei. This Caspase 1 antibody generated a diffused but specific cytoplasmic staining in columnar epithelia cells of villi, and a few cells depicted nuclear staining also. Only a subset of connective tissue cells in lamina propria depicted positivity (cytoplasmic) for this protein.

Immunocytochemistry/Immunofluorescence: IRE1 alpha Antibody [NB100-2324] - IRE-1 alpha antibody was tested in HeLa cells at 1:200 against Dylight 488 (Green). Alpha tubulin and nuclei were counterstained against Dylight 568 (Red) and DAPI (Blue).

Immunohistochemistry-Paraffin: IRE1 alpha Antibody [NB100-2324] - Analysis of a formalin fixed mouse colon tissue section using total IRE1 alpha antibody (NB100-2324) at 1:300. Note a strong cytoplasmic signal in various cells of colonic villi (columnar epithelial and goblet cells) and the muscular layer. A few cells, potentially with active UPR, showed nuclear positivity also while the signal was negligible in cells at base of crypts and lamina propria.

Flow Cytometry: NQO-1 Antibody (A180) [NB200-209] - An intracellular stain was performed on U-87 cells with NQO-1 [A180] Antibody NB200-209AF647 (blue) and a matched isotype control (orange). Cells were fixed with 4% PFA and then permeabilized with 0.1% saponin. Cells were incubated in an antibody dilution of 2.5 ug/mL for 30 minutes at room temperature. Both antibodies were conjugated to Alexa Fluor 647.

Flow (Intracellular): NQO-1 Antibody (A180) [NB200-209] - An intracellular stain was performed on U87MG cells with NQO1 (A180) antibody NBP2-24917 (blue) along with a matched isotype control NBP2-27287 (orange). Cells were fixed with 4% PFA and permeabilized with 0.1% saponin. Cells were incubated in an antibody dilution of 1 ug/mL for 30 minutes at room temperature, followed by mouse F(ab)2 IgG (H+L) APC-conjugated secondary antibody (F0101B, R&D Systems).

Flow Cytometry: NQO-1 Antibody (A180) [NB200-209] - An intracellular stain was performed on U-87 cells with NQO-1 [A180] Antibody NB200-209C (blue) and a matched isotype control (orange). NQO-1 (A180) was detected in immersion fixed U-2 OS human osteosarcoma cell line using Mouse anti-NQO-1 (A180) Protein G Purified Monoclonal Antibody conjugated to DyLight 650 (Catalog # NB200-209C) (light blue) at 10 µg/mL overnight at 4C. Cells were counterstained with DAPI (blue). Cells were imaged using a 100X objective and digitally deconvolved.

Immunohistochemistry-Paraffin: SR-BI Antibody [NB400-113] - Analysis of a FFPE section of human adrenal gland tissue using 1:100 dilution of SR-BI antibody. The staining was developed using HRP-DAB based detection method and the nuclei of the cells were counterstained with hematoxylin. This antibody generated a specific staining of SR-BI/SCARB1 in the glandular cells. The staining was membrane-cytoplasmic in the zona glomerulosa cells while the signal was primarily localized to the membranes of the cells in the zona fasciculata and zona reticularis layers of the adrenal cortex.

Immunocytochemistry/Immunofluorescence: Apolipoprotein E/ApoE Antibody (WUE-4) [NB110-60531] - HepG2 cells were fixed for 10 minutes using 10% formalin and then permeabilized for 5 minutes using 1X TBS + 0.5% Triton X-100. The cells were incubated with anti-ApoE (WUE-4) [NB110-60531] at a 1:200 dilution overnight at 4C and detected with an anti-mouse DyLight 488 (Green) at a 1:500 dilution. Actin was detected with Phalloidin 568 (Red) at a 1:200 dilution. Nuclei were counterstained with DAPI (Blue). Cells were imaged using a 40X objective.

Flow Cytometry: iNOS Antibody (4E5) [NBP2-22119] - An intracellular stain was performed on A549 cells with NBP2-22119APC (blue) and a matched isotype control (orange). Cells were fixed with 4% PFA and then permeabilized with 0.1% saponin. Cells were incubated with antibody at 1 ug/mL for 30 minutes at room temperature. Both antibodies were conjugated to allophycocyanin (APC).

Immunofluorescence (IF) of proteins identified by LCM/MS (A–D) “expected” or (E–H) “not expected” to be within protrusions based on their known functions/localizations. (A) Cd47 & (B) Anxa2 are found in both hCAD/dCAD protrusions. (C) Tenm2 is found within hCAD protrusions & (D) Cobl in GCs.
