![Immunohistochemistry-Paraffin: Mouse Monoclonal alpha-Smooth Muscle Actin Antibody (1A4/asm-1) [NBP2-33006] - Images demonstrating SMA immunoreactivity in a variety of horse FFPE tissue sections. NBP2-33006 was used at a concentration of 125ng/mL and was left on tissue sections for 30m at room temperature. Formalin fixed paraffin embedded sections were deparaffinized and rehydrated. Sections then underwent heat induced epitope retrieval in a citrate-based solution for 5min in a food steamer. Endogenous peroxidase activity was blocked by incubating slides for 15min in tris buffered saline containing 3% (volume/volume) hydrogen peroxide. Tissue sections were then washed, blocked with normal horse serum for 20 min, and then incubated with the primary antibody (125ng/mL) for 30min at room temperature. Sections were then washed and incubated with a horse anti-mouse HRP polymer for 30m at room temperature. Sections were washed and incubated with DAB chromogen for approximately 2.5min. Sections were then washed, counterstained with hematoxylin, dehydrated, cleared and mounted with a coverslip. This antibody generated a specific staining of SR-BI/SCARB1 in the glandular cells. ![Gel Super Shift Assays: HIF-1 alpha Antibody [NB100-654] - Mouse HIF-1 alpha+/+ and HIF-1 alpha+/- cells were exposed to hypoxia or 125 uM CoCl2 for 6 h, nuclear extracts were then prepared and EMSA carried out. HIF-1 isolated from hypoxia or CoCl2-treated cells bound to the wild-type probe (lanes #1-6) but not to the mutant probe (lanes #7-12). HIF-1/DNA complex was detected in hypoxia (H)-or CoCl2 (Co)-treated cells (lanes #2, 3, 5, 6) but not in control (C) cells (lanes #1, 4). More complex (darker band) was seen in hypoxia-or CoCl2-treated HIF-1 alpha+/+ cells (lanes #2, 3) than that in hypoxia-or CoCl2-treated HIF-1 alpha+/- cells (lanes #5, 6). Immortalized human mesenchymal stem cells were differentiated under osteogenic conditions (see Materials and methods) and assayed for changes in LC3I and LC3II during a 72-hour window. Cells were differentiated under standard conditions (top) or with addition of 5 uM rapamycin (middle) or 5 nM bafilomycin (bottom) for the first 3 hours of differentiation to modulate autophagy. Immunoblots were performed for LC3 at the indicated time points to assess autophagosome degradation via relative changes in LC3II (lower band; 17 kDa). Studies were repeated three times with similar trends seen consistently. ![Western Blot: CD63 Antibody (MEM-259) - BSA Free [NB100-77913] - Cytokine & checkpoint molecule arrays from plasma exosomes showed a decreased concentration of IFN-gamma , IL-10, IL-13, B7-1, B7-2, & ICOSL in GBM patients in comparison to normal donors. (A) Quantification of IFN-gamma , IL-10, & IL-13 in plasma exosomes from normal donors & grade 4 GBM patients' plasma exosomes (mean ± SEM, n = 4/group, *P < 0.05). (B) Quantification of B7-1, B7-2, & ICOSL in plasma exosomes from normal donors & grade 4 GBM patients' plasma exosomes (mean ± SEM, n = 4/group). (C) Quantification of the immunosuppressive checkpoint & T cell costimulatory homolog protein PD-L1 in plasma exosomes from normal donors & grade 4 GBM patients' plasma exosomes (mean ± SEM, n = 4/group). Interestingly, PD-L1 is found at similar levels in both normal donor & glioma patient plasma exosomes & these findings were confirmed by western blot. Western Blot analysis also shows that exosomal markers Flotillin-1 & CD63 are found universally in plasma exosomes from normal donors (n = 3), glioblastoma (grade 4) patients (n = 4), & grade 3 glioma patients (n = 3), though CD63 expression may be slightly reduced in glioma patients.
