Immunocytochemistry/Immunofluorescence: Fibronectin Antibody - BSA Free [NBP1-91258] - NIH-3T3 cells were fixed for 10 minutes using 10% formalin and then permeabilized for 5 minutes using 1X TBS + 0.5% Triton X-100. The cells were incubated with anti-Fibronectin at 2 ug/mL overnight at 4C and detected with an anti-rabbit DyLight 488 (Green) at 1:500. Alpha tubulin (DM1A) NB100-690 was used as a co-stain at 1:1000 and detected with an anti-mouse DyLight 550 (Red) at a 1:500 dilution. Nuclei were counterstained with DAPI (Blue). Cells were imaged using a 40X objective.

Fibronectin was detected in immersion fixed HepG2 human hepatocellular carcinoma cell line using Rabbit anti-Fibronectin Affinity Purified Polyclonal Antibody conjugated to Alexa Fluor® 647 (Catalog # NBP1-91258AF647) (light blue) at 10 µg/mL overnight at 4C. NBP2-33006 was used at a concentration of 125ng/mL and was left on tissue sections for 30m at room temperature. Formalin fixed paraffin embedded sections were deparaffinized and rehydrated. Sections then underwent heat induced epitope retrieval in a citrate-based solution for 5min in a food steamer. Endogenous peroxidase activity was blocked by incubating slides for 15min in tris buffered saline containing 3% (volume/volume) hydrogen peroxide. Tissue sections were then washed, blocked with normal horse serum for 20 min, and then incubated with the primary antibody (125ng/mL) for 30min at room temperature. Sections were then washed and incubated with a horse anti-mouse HRP polymer for 30m at room temperature. Sections were washed and incubated with DAB chromogen for approximately 2.5min. <b>Genetic Strategies Validation. </b>Western Blot: EGLN3/PHD3 Antibody [NB100-139] - BxPC3 cells stably transduced with retrovirus containing PHD3Wt (BxPC3-Wt), Vector (BxPC3-Vec) or anti-PHD3 shRNA (BxPC3-KD) were harvested for RNA and protein following 24 hours exposure to normoxia (21% O2) or hypoxia (1% O2). Whole cell lysate from BxPC3-Wt, BxPC3-Vec and BxPC3-KD cells following 24 hours exposure to normoxia (N) or hypoxia (H) was resolved by SDS-PAGE and blotted for actin (top) and PHD3 (bottom). PHD3 band intensity was quantified relative to actin in each lane and then normalized to BxPC3-Vec (N). Relative band intensity is indicated below the figure. HCT116 & DLD-1 cells were cultured in DMEM for 6, 24 & 48 h either in the absence or in the presence of 5-dAzaC at a concentration of 1.00 or 5.00 μM under hypoxic or normoxic conditions. After incubation the cells were used for total RNA isolation & protein isolation. Total RNA was reverse-transcribed, & PHD3 cDNA levels were determined by RQ-PCR relative quantification analysis. RQ-PCR results were standardized by the geometric mean of PBGD & hMRPL19 cDNA levels. PHD3 cDNA levels are expressed as a multiplicity of the respective controls. Each sample was determined in triplicate & results are presented as the mean ± SE from three experiments *P < 0.05. The cell protein was separated by 10% SDS-PAGE, & transferred to a membrane that was then immunoblotted with Rp anti - PHD3 Ab & incubated with goat anti-rabbit HRP-conjugated Ab. The membrane was then stripped & reblotted with Rp anti-GAPDH Ab, followed by incubation with goat anti-rabbit HRP-conjugated Ab. The band densitometry readings were normalized to GAPDH loading control. The ratio PHD3 to GAPDH for control was assumed to be 1. CD31/PECAM-1 was detected in immersion fixed THP-1 (red, positive) but not THP-1 KO (negative) cells using 8 ug/mL Mouse Anti-Human CD31/PECAM-1 (JC/70A) Monoclonal Antibody (Catalog # NB600-562) for 3 hours at room temperature. Cells were stained with the NorthernLights(TM) 557-conjugated Donkey anti-Mouse IgG Secondary Antibody (red, Catalog # NL007)) and and counterstained with DAPI (blue).

<b>Genetic Strategies Validation and Orthogonal Strategies Validation.</b>Formalin-fixed paraffin-embedded tissue sections of human metastatic tonsil were probed for CD31 mRNA (ACD RNAScope probe, catalog # 487381; Fast Red chromogen, ACD catalog # 322500). Adjacent tissue section was processes for immunohistochemistry using mouse monoclonal (NB600-562) at 1:25 dilution for 1 hour at room temperature followed by incubation with the anti-mouse IgG VisUCyte HRP Polymer Antibody (Catalog # VC001) and DAB chromogen (yellow-brown). A surface stain was performed on Jurkat cells with the Dylight 550 conjugate of CD31/PECAM-1 Antibody (JC/70A) (NB600-562R, blue) and a matched isotype control (orange). Cells were incubated in an antibody dilution of 5 ug/mL for 20 minutes at room temperature. Both antibodies were conjugated to DyLight 550.

HUVEC cells were fixed for 10 minutes using 10% formalin and then permeabilized for 5 minutes using 1X TBS + 0.5% Triton X-100. The cells were incubated with CD31/PECAM-1 Antibody (JC/70A) at 2 ug/ml overnight at 4C and detected with an anti-mouse DyLight 488 (Green) at a 1:500 dilution. Actin was detected with Phalloidin 568 (Red) at a 1:200 dilution. Nuclei were counterstained with DAPI (Blue). Cells were imaged using a 40X objective.

NQO-1 (A180) was detected in immersion fixed U-2 OS human osteosarcoma cell line using Mouse anti-NQO-1 (A180) Protein G Purified Monoclonal Antibody conjugated to DyLight 650 (Catalog # NB200-209C) (light blue) at 10 µg/mL overnight at 4C. Cells were counterstained with DAPI (blue). Cells were imaged using a 100X objective and digitally deconvolved.

Western Blot: GAPDH Antibody [NB300-327] - Western blot analysis of extracts from Jurkat and HeLa cells using NB300-327 at 1:1000. Theoretical molecular weight: 36 kDa.

$Western Blot: GAPDH Antibody [NB300-327] - Analysis of different cell cytosolic or nuclear enriched fractions, GAPDH antibody, dilution 1:20,000 (Red): [1] protein standard, [2] NIH-3T3 cytosolic, [3] NIH-3T3 nuclear, [4] HeLa cytosolic, and [5] HeLa nuclear fractions. Strong band at 37kDa corresponds to GAPDH protein, mainly detected in the cytosolic fractions. The same blot was simultaneously probed with mouse mAb to the nuclear RNA binding protein SF3B4, dilution 1:1,000 (Green). In contrast to GAPDH, SF3B4 is exclusively expressed in the nuclear fraction.$

<b>Biological Strategies Validation. </b>Immunohistochemistry: ASC/TMS1 Antibody [NBP1-78977] - Rats were infected by S. pneumoniae or given PBS sham control. 21.5 umoles/kg Mito-Vit-E (MVE) or vehicle was administered orally 30 minutes post-inoculation, and heart tissues were harvested 24 hours later. Heart sections were co-stained with anti-ASC (brown) and haematoxylin (blue). Negative control was stained with secondary antibody alone. Western Blot: ASC/TMS1 Antibody [NBP1-78977] - Whole cell protein from (1) THP1 and (2) HL60 was separated on a 4-15% gel by SDS-PAGE, transferred to 0.2 um PVDF membrane and blocked in 5% non-fat milk in TBST. The membrane was probed with 4.0 ug/ml anti-ACS/TMS1 in 1% milk, and detected with an anti-rabbit HRP secondary antibody using chemiluminescence.

Flow Cytometry: ASC/TMS1 Antibody - BSA Free [NBP1-78977] - An intracellular stain was performed on MCF7 cells with ASC/TMS1 NBP1-78977 (blue) and a matched isotype control NBP2-24891 (orange). Cells were fixed with 4% PFA and then permeabilized with 0.1% saponin. Cells were incubated in an antibody dilution of 1 ug/mL for 30 minutes at room temperature, followed by Rabbit IgG (H+L) Cross-Adsorbed Secondary Antibody, Dylight 550 (SA5-10033, Thermo Fisher).

Immunocytochemistry/Immunofluorescence: ASC/TMS1 Antibody [NBP1-78977] - Tested in MCF-7 cells with FITC (green). Nuclei and alpha-tubulin were counterstained with DAPI (blue) and DyLight 550 (red).

Immunocytochemistry/Immunofluorescence: S100A7/Psoriasin Antibody (47C1068) [NB100-56559] - S100A7/psoriasin detection was performed on (A) untreated and (B) LPS-treated (100 ng/mL, 21 hrs) HOPE-fixed NCI-H727 cells using the monoclonal Psoriasin/HID5/S100A7 antibody (NB100-56559). (C) Positive control is shown by the expression of the nuclear Ki67 antigen with the MIB-1 (2 ug/mL) antibody. (D) Negative control was included omitting the primary antibody.

Cells were fixed with 4% PFA and then permeablized with 0.1% saponin. Cells were incubated in an antibody dilution of 1 ug/mL for 30 minutes at room temperature. Diffused to granular cytoplasmic immunostaining was observed all over the tissue section with highest expression levels in the areas with cancer cells especially in the developing squamous keratin pearls.
