WNT5A: wingless-type MMTV integration site family, member 5A. Entrez Protein: NP_003383. The WNT gene family consists of structurally related genes which encode secreted signaling proteins. These proteins have been implicated in oncogenesis and in several developmental processes, including regulation of cell fate and patterning during embryogenesis. This gene is a member of the WNT gene family. It encodes a protein which shows 98%, 98% and 87% amino acid identity to the mouse, rat and the xenopus Wnt5A protein, respectively. The experiments performed in Xenopus laevis embryos identified that human frizzled-5 (hFz5) is the receptor for the Wnt5A ligand and the Wnt5A/hFz5 signaling mediates axis induction.

I notice that the Western blot analysis is using WNT5A-hIigGFc transfected HEK293 cell lysate. Is the hIigGFc increasing the molecular weight of WNT5A? And what is the molecular weight of WNT5A-hligGFc?
The hlgGFc is a tag used for purification purposes. The molecular weight of the WNT5A-hlgGFc complex is around 65.2 kDa
I have one question about your product of WNT5A Antibody (NBP1-47438) before I decide to buy it. The TMW of WNT5A is 42.3 in the instruction, but the Western blot photo shows it between 72kDa and 55kDa. Can you tell me the reason and give me some advice?
WNT5A has multiple glycosylation sites (UniProt P41221). Because of this, WNT5A will run on the gel at a higher apparent molecular weight than the predicted 42.3 kDa.
I need to block NBP1-47438 with Wnt5a peptide to see the specificity of the antibody. Do you have any suggestions?
In our blocking experiments with unpurified antibodies we use a 4-5ug of blocking agent/1ul of antibody. This generally yields good results. I would recommend doing a titre to ensure ample blocking of the signal.
It is WNT5A antibody 3D10. I wonder how was Western blot performed with this antibody. Was this a native gel, non-reducing conditions? Why is the band at 70kDa when the theoretical MW is 43kDa? Is it because of the tag?
The increased molecular weight is due to the human Fc tag. We used our general WB protocol while working with this antibody.
