Samples: Whole cell lysate (1.0 mg per IP reaction; 20% of IP loaded) from MCF-7 cells prepared using NETN lysis buffer. Antibodies: Affinity purified rabbit anti-UBQLN1 antibody (NBP3-29774 lot 1) used for IP at 1 µg per reaction. UBQLN1 was also immunoprecipitated by rabbit anti-UBQLN1 antibody BL20647. For blotting immunoprecipitated UBQLN1, NBP3-29774 was used at 0.04 mg/ml. Detection: Chemiluminescence with an exposure time of 30 seconds.

Background

Ubiquilin 1 encodes an ubiquitin-like protein (ubiquilin) that shares high degree of similarity with related products in yeast, rat and frog. Ubiquilins contain a N-terminal ubiquitin-like domain and a C-terminal ubiquitin-associated domain. They physically associate with both proteasomes and ubiquitin ligases, and thus are thought to functionally link the ubiquitination machinery to the proteasome to affect in vivo protein degradation. This ubiquilin has also been shown to modulate accumulation of presenilin proteins, and is found in lesions associated with Alzheimer's and Parkinson's disease. Two transcript variants encoding different isoforms have been found for this gene.
