Western Blot: Syntaxin-BP1 Antibody (S05-4B6) [NBP3-15097] - Western blot detection of Syntaxin-BP1 in Hela, A549, C6 cell lysates using Syntaxin-BP1 Rabbit mAb (1:1000 diluted). Predicted band size: 68kDa. Observed band size: 68kDa.

Western blot shows lysates of U‑87 MG human glioblastoma/astrocytoma parental cell line and Syntaxin-BP1 knockout U-87 MG cell line (KO). Nitrocellulose membrane was probed with Syntaxin-BP1 Antibody (S05-4B6) (Catalog # NBP3-15097) followed by HRP-conjugated secondary antibody. A specific band was detected for Syntaxin-BP1 at approximately 68 kDa (as indicated) in the parental U-87 MG cell line, but is not detectable in knockout U-87 MG cell line. U‑87 MG human glioblastoma/astrocytoma cell line lysates were prepared and immunoprecipitation was performed using 2.0 μg of Syntaxin-BP1 Antibody (S05-4B6) (Catalog # NBP3-15097) pre-coupled to Dynabeads Protein A. Immunoprecipitated Syntaxin-BP1 was detected in Western Blot with NBP3-15097 used at 1/2000. The Ponceau stained transfer of the blot is shown. U‑87 MG human glioblastoma/astrocytoma parental cell line WT and Syntaxin-BP1 U-87 MG KO cells were labelled with a green or a far-red fluorescent dye, respectively. Cells were stained with Syntaxin-BP1 Antibody (S05-4B6) (Catalog # NBP3-15097) followed by incubation with an Alexa-fluor 555 conjugated secondary antibody (upper panel). DAPI-only counterstained cells shown on a lower panel. Acquisition of the blue (nucleus-DAPI), green (identification of WT cells), red (antibody staining) and far-red (identification of KO cells) channels was performed. Background

Munc-18 (mammalian homologue of Unc-18) is a protein that is thought to be involved in regulating exocytosis due, at least in part to its ability to bind syntaxin (Ciufo et al., 2005). Munc18-1 is a neuron-specific member of the Sec1/Munc18 protein family that binds to syntaxin1A and is thought to stabilize the complex (Liu et al., 2004). The function of Munc-18 is thought to be regulated by PKC phosphorylation of Ser515 on the Munc-18 protein (Sassa et al., 1996).
