Western Blot: STAG2 Antibody [NB100-301] - Samples: Whole cell lysate (50 ug) from HeLa, 293T, and mouse NIH3T3 cells prepared using NETN lysis buffer. Antibody : Affinity purified rabbit anti-SA2 antibody used for WB at 0.1 ug/ml. Detection: Chemiluminescence with an exposure time of 10 seconds.

Immunocytochemistry/Immunofluorescence: STAG2 Antibody [NB100-301] - HeLa cells that were extracted for 5 min. at 4C in 0.5% Triton in CSK buffer. Antibody used at 1 ug/ml.

Immunoprecipitation: STAG2 Antibody [NB100-301] - Whole cell lysate (0.5 or 1.0 mg per IP reaction; 20% of IP loaded) from HeLa cells prepared using NETN lysis buffer. Antibodies: Affinity purified rabbit anti-SA2 antibody used for IP at 6 ug per reaction. SA2 was also immunoprecipitated by rabbit anti-SA2 antibodies. For blotting immunoprecipitated SA2,was used at 1 ug/ml. Detection: Chemiluminescence with an exposure time of 10 seconds.

Background

SA2 is a component of the cohesin complex, which is required for the cohesion of sister chromatids after DNA replication. The cohesin complex apparently forms a large proteinaceous ring within which, sister chromatids can be trapped. At anaphase, the complex is cleaved and dissociates from chromatin, allowing sister chromatids to segregate. The cohesin complex may also play a role in spindle pole assembly during mitosis. The subunit interacts directly with RAD21 in cohesin complex. Cohesin complexes are composed of a heterodimer between a SMC1 protein (SMC1L1 or SMC1L2) and SMC3, which are attached via their hinge domain, and RAD21 links them at their heads, and one STAG protein (STAG1, STAG2 or STAG3). In cohesin complexes, STAG2 is mutually exclusive with STAG1 and STAG3.
