Src Antibody Pack

Western blot shows lysates of MDA-MB-468 human breast cancer cell line and HepG2 human hepatocellular carcinoma cell line untreated (-) or treated (+) with 50 µM pervanadate (PV) for 10 minutes. PVDF membrane was probed with 1 µg/mL of Rabbit Anti-Human Phospho-Src (Y419) Antigen Affinity-purified Polyclonal Antibody (Catalog # AF2685), followed by HRP-conjugated Anti-Rabbit IgG Secondary Antibody (Catalog # HAF008). A specific band was detected for Phospho-Src (Y419) at approximately 56 - 61 kDa (as indicated). This experiment was conducted under reducing conditions and using Immunoblot Buffer Group 1.

A specific band was detected for Src at approximately 60 kDa (as indicated). This experiment was conducted under reducing conditions and using Immunoblot Buffer Group 1.

A specific band was detected for Src at approximately 60 kDa (as indicated). This experiment was conducted under reducing conditions and using Immunoblot Buffer Group 1.

Background

Src is a protein tyrosine kinase known to regulate cellular adhesion. Composed of three major domains; SH2, SH3 and a kinase catalytic domain, Src can be switched from an inactive to an active state though control of its phosphorylation state or through protein-protein interaction. Phosphorylation of Y529 by Csk and Chk inactivates Src, while dephosphorylation of Y529 will modify its conformation to an open active state (1-3). Mutation of Y529 leads to Scr overactivity. Several cancers including colon and breast cancer have been associated with an increase of Src activity (4).
