Background
Sir2 (silent information regulator 2) protein plays a crucial role in regulating the life span of C. elegans by means of caloric restricion. Overexpression of the protein can extend the life span, and is, thus, a regulator of longevity.

Works well on blot with many different ECL preps. Originally, there was quite a lot of background signal with this Ab, mostly from IgG. I was able to greatly mitigate this by using Clean-Blot IP detection reagent (HRP) from Thermo Scientific, which binds native IgG with a much higher affinity versus denatured.

NB100-1923 is used to detect the C. elegans protein; yeast is not a validated species. The species used in the western blot was C. elegans.
