This affinity purified antibody has been tested for use in ELISA, western blot, and IHC. This antibody is suitable for immunofluorescence. Specific conditions for reactivity should be optimized by the end user. Expect a band approximately 37 kDa in size corresponding to Sfrp1 by western blotting in the appropriate cell lysate or extract. |
Reactivity Notes
A BLAST analysis was used to suggest reactivity with this protein from human based on 100% homology for the immunogen sequence. Expect cross reactivity with sFRP-1 from mouse, rat and bovine sources as only a single amino acid residue change is found within the immunogen sequence (91% positive by BLAST). Cross reactivity with sFRP-1 homologues from other sources has not been determined.

Background
SFRP1 (also known as FRP, FRP1, SARP2, Secreted Apoptosis-related Protein 2, Secreted Frizzledrelated Protein and Secreted frizzled-related protein 1) is a member of the SFRP family that contains a cysteine-rich domain homologous to the putative Wnt-binding site of Frizzled proteins. SFRPs act as soluble modulators of Wnt signaling. SFRP1 and SFRP5 may be involved in determining the polarity of photoreceptor cells in the retina. SFRP1 is expressed in several human tissues, with the highest levels in heart.

May I request confirmation that SFRP1 Antibody (NB600-499) has been tested in ELISA using cell lysate as sample instead of immunogen as sample? Will this antibody detect endogenous levels of the protein in ELISA? We have a customer who has had sensitivity issues with other SFRP1 antibodies and is looking for one that can detect endogenous levels of the protein. Thanks for your help. (This product lists ELISA as an application, what samples were used and can the endogenous protein be detected by ELISA?)
- I have pulled up information on this product and it looks like it has been tested in ELISA on the immunogen as opposed to an endogenous cell lysate. However, we do have a guarantee policy and we have confirmed this antibody to detect properly in Western Blot with cell lysate containing endogenous levels of the protein so it should also have an affinity for the protein in ELISA as well. (We have tested this SFRP1 antibody in ELISA using the native full length protein. While we have confirmed the presence of endogenous protein in cell lysate samples via western blot, we have not used cell lysate samples for detection in ELISA. Due to the western blot results, we predict that cell lysates containing endogenous levels of the SFRP1 protein should bind and detect the antibody.)
