• Application Notes: Western blot (1:500 - 1:1,000) and electrophoretic mobility shift assay (EMSA). For Western blot, a 44 kDa band should be detected. For optimal results, primary antibody incubations should be performed at room temperature. The addition of 0.1% Tween 20 to all Blotto solutions may also reduce background. Further optimization may be required by the end user.

Background

RUNX3 binds to the core site, 5'-PYGPYGGT-3', of a number of enhancers and promoters, including murine leukemia virus, polyomavirus enhancer, T-cell receptor enhancers, lck, IL-3 and GM-CSF promoters. RUNX3 consists of a heterodimer of an alpha and a beta subunit. The alpha subunit binds DNA as a monomer through the Runt domain. DNA-binding is increased by heterodimerization.
