Western Blot: RSK1 [p Thr573] Antibody (JE50-12) [NBP3-32798] - Western blot analysis of RSK1 on different lysates with Rabbit anti-RSK1 antibody (NBP3-32798) at 1/1,000 dilution. Lane 1: K-562 cell lysate Lane 2: K-562 treated with 500nM PMA for 15 minutes cell lysate Lane 3: HeLa cell lysate Lane 4: HeLa treated with 200nM PMA for 4 hours cell lysate Lysates/proteins at 20 ug/Lane. Predicted band size: 83 kDa Observed band size: 83 kDa Exposure time: 1 minute; 4-20% SDS-PAGE gel. Proteins were transferred to a PVDF membrane and blocked with 5% NFDM/TBST for 1 hour at room temperature. The primary antibody (NBP3-32798) at 1/1,000 dilution was used in 5% NFDM/TBST at 4C overnight. Goat Anti-Rabbit IgG - HRP Secondary Antibody at 1/50,000 dilution was used for 1 hour at room temperature.

Background

Rsk-1 (p90-Rsk-1) is a member of a family of 90 kDa ribosomal protein S6 kinases, which includes Rsk-1, Rsk-2 and Rsk-3. These are broadly expressed serine/threonine protein kinases (1) activated in response to mitogenic stimuli, including extracellular signal-regulated protein kinases Erk1 and Erk2 (2). p90-Rsk-1 is activated by MAPK in vitro and in vivo via phosphorylation (3). Active Rsks appear to play a major role in transcriptional regulation by translocating to the nucleus and phosphorylating c-Fos and CREB (2,4).
