RNF8 Antibody (JE56-43)

Western Blot: RNF8 Antibody (JE56-43) [NBP3-32912] - Western blot analysis of RNF8 on human brain tissue lysates with Rabbit anti-RNF8 antibody (NBP3-32912) at 1/2,000 dilution. Lysates/proteins at 40 ug/Lane. Predicted band size: 56 kDa Observed band size: 63 kDa Exposure time: 3 minutes; 4-20% SDS-PAGE gel. Proteins were transferred to a PVDF membrane and blocked with 5% NFDM/TBST for 1 hour at room temperature. The primary antibody (NBP3-32912) at 1/2,000 dilution was used in 5% NFDM/TBST at 4C overnight. Goat Anti-Rabbit IgG - HRP Secondary Antibody at 1/50,000 dilution was used for 1 hour at room temperature.

Immunohistochemistry: RNF8 Antibody (JE56-43) [NBP3-32912] - Immunohistochemical analysis of paraffin-embedded human brain tissue with Rabbit anti-RNF8 antibody (NBP3-32912) at 1/500 dilution. The section was pre-treated using heat mediated antigen retrieval with sodium citrate buffer (pH 6.0) for 2 minutes. The tissues were blocked in 1% BSA for 20 minutes at room temperature, washed with ddH2O and PBS, and then probed with the primary antibody (NBP3-32912) at 1/500 dilution for 1 hour at room temperature. The detection was performed using an HRP conjugated compact polymer system. DAB was used as the chromogen. Tissues were counterstained with hematoxylin and mounted with DPX.

Background

RNF8 is encoded by this gene contains a RING finger motif and a FHA domain. This protein has been shown to interact with several class II ubiquitin-conjugating enzymes (E2), including UBE2E1/UBCH6, UBE2E2, and UBE2E3, and may act as an ubiquitin ligase (E3) in the ubiquitination of certain nuclear proteins. Alternatively spliced transcript variants encoding distinct isoforms have been reported.
