Background

Recombinant human Collagen XV alpha 1/COL15A1 is the C-terminal endostatin-like domain (amino acids 1133-1381) of human Collagen XV. Collagen XV is one of the two members of a vertebrate collagen family termed multiplexins. The other member of this family is Collagen XVIII. This family is characterized by a collagen triple helix region that is interrupted by several non-collagenous stretches, as well as a C-terminal non-triple helical NC1 domain (1, 2). The NC1 domain is composed of three functionally distinct regions: an association region in its most N-terminal part that trimerizes NC1 monomers, a protease-sensitive hinge region, and the C-terminal globular endostatin (ES) domain, or in the case of Collagen XV, an ES-like domain (2, 3). Both Collagen XV and Collagen XVIII are located in the basement membrane or the basement zone. They show anti-angiogenic and anti-tumoral properties (4), and play an important role in tissue homeostasis in liver, kidney, eye and the central nervous system (5). The Drosophila multiplexin (Dmp) is shown to modulate axon pathfinding accuracy in motor pathways (1). Within amino acids 1133-1381, the human protein shares 74% sequence homology with its mouse counterpart, and 73% sequence homology with that of the rat. This ES-like domain in Collagen XV show approximately 60% sequence identity with the endostatin domain of type XVIII Collagen. Studies have shown that endostatins derived from Collagens XV and XVIII differ in structural and binding properties, tissue distribution and anti-angiogenic activity (6).
