Background

PTPRB is a member of the protein tyrosine phosphatase (PTP) family. The family comprises at least 37 proteins, characterized by a catalytic phosphatase domain of approximately 240 amino acids, and includes both transmembrane and cytosolic enzymes. PTP1B is a transmembrane PTP. The PTPs have high substrate specificity for phosphotyrosyl proteins, at the primary sequence level sharing little similarity with the protein serine phosphatases, protein threonine phosphatases, or the acid and alkaline phosphatases. PTP beta interacts with and regulates the tyrosine phosphorylation level of catenins, which are critical in physiological and pathological events such as cell migration, adhesion and transformation.

Immunocytochemistry/Immunofluorescence: Tenascin C Antibody (4C8MS) [NB110-68136] - SK-MEL-28 cells were fixed for 10 minutes using 10% formalin and then permeabilized for 5 minutes using 1X PBS + 0.05% Triton-X100. The cells were incubated with anti-Tenascin C at 5 ug/ml overnight at 4C and detected with an anti-Mouse IgG Dylight 488 (Green) at a 1:500 dilution. Nuclei were counterstained with DAPI (Blue). Cells were imaged using a 40X objective.

PVDF membrane was probed with 1.0 ug/ml of Mouse Anti-Human NG2/MCSP (LHM2) Polyclonal Antibody (Catalog # NB100-2688) followed by HRP-conjugated Anti-Mouse IgG Secondary Antibody (Catalog #HAF018). Specific band was detected for NG2/MCSP (LHM2) at approximately 300 kDa (as indicated) in the parental HeLa cell line, but is not detectable in the knockout HeLa cell line. This experiment was conducted under reducing conditions.

The GFAP antibody stains the network of astrocytic cells and the processes of Bergmann glia in the molecular layer. The MeCP2 antibody specifically labels nuclei of certain neurons.
Western Blot: GFAP Antibody [NB300-141] - Analysis of different tissue lysates using rabbit polyclonal antibody to GFAP, NB300-141, dilution 1:5000 in green: [1] protein standard (red), [2] rat brain, [3] rat spinal cord, [4] mouse brain, [5] mouse spinal cord. Strong band at about 50 kDa corresponds to the major isotype of the GFAP protein. Smaller isotypes and proteolytic fragments of GFAP are also detected on the blot.
Western Blot: GFAP Antibody [NB300-141] - Analysis of Rat brain lysate. Antibody at 1:5000. Specific band running with an apparent SDS-PAGE molecular weight of ~50 kDa corresponds to rodent GFAP was observed.
