Background

Podoplanin(PDPN) is a small mucin-like transmembrane protein, widely expressed in various specialized cell types throughout the body. This is a type-I integral membrane glycoprotein with diverse distribution in human tissues. The physiological function of this protein may be related to its mucin-type character. The homologous protein in other species has been described as a differentiation antigen and influenza-virus receptor. Recombinant human PDPN protein, fused to His-tag at N-terminus, was expressed in E.coli and purified by conventional chromatography techniques.

  1. What is the procedure to denature podoplanin protein?
    • We denature our proteins by diluting them with loading buffer and boiling them at 95 degrees Celsius for 5-10 minutes. If reducing conditions are desired, we add 5% BME or DTT to the loading buffer prior to boiling the sample. With larger proteins (+200kDa) we will heat the samples at 65 degrees Celsius for 10 minutes.
