Background

Human PLZF is a 74 kDa nuclear protein that belongs to the POK family of transcriptional repressors. It is a 673 amino acid protein that contains an N-terminal BTB domain, followed by an acidic domain, a proline-rich region and a C-terminal zinc-finger domain. PLZF forms homodimers with RAR alpha and LAZ3 with its zinc-finger region. Alternate splice forms exist which are tissue-specific and show a deletion of either the BTB domain, the acidic region, or the proline-rich region. Human PLZF shares 96%, 97%, 96%, and 96% amino acid identity with rat, mouse, canine, and bovine PLZF, respectively.

Immunohistochemistry-Paraffin: PML Protein Antibody [NB100-59787] - IHC analysis of a formalin fixed and paraffin embedded tissue section of human stomach using 1:50 dilution of PML antibody. The signal was developed using HRP-labeled secondary with DAB reagent followed by hematoxylin counterstaining. This antibody generated an expected strong nuclear staining in most of the cells in the glandular as well as the connective tissue. The cells showed some cytoplasmic signal also which was much weaker than the nuclear staining.

PVDF membrane was probed with 2.0 ug/ml of Rabbit Anti-Human HDAC1 Polyclonal Antibody (Catalog # NB100-56340) followed by HRP-conjugated Anti-Rabbit IgG Secondary Antibody (Catalog #HAF008). Specific band was detected for HDAC1 at approximately 65 kDa (as indicated) in the parental HeLa cell line, but is not detectable in the knockout HeLa cell line. This experiment was conducted under reducing conditions.

Immunohistochemistry-Paraffin: HDAC1 Antibody [NB100-56340] - Human testis using HDAC1 antibody at 1:250 on a Bond Rx autostainer (Leica Biosystems). The assay involved 20 minutes of heat induced antigen retrieval (HIER) using 10mM sodium citrate buffer (pH 6.0) and endogenous peroxidase quenching with peroxide block. The sections were incubated with primary antibody for 30 minutes and Bond Polymer Refine Detection (Leica Biosystems) with DAB was used for signal development followed by counterstaining with hematoxylin. Whole slide scanning and capturing of representative images was performed using Aperio AT2 (Leica Biosystems). Staining was performed by Histowiz.
