• Immunogen: Purified recombinant human platelet-derived growth factor (PDGF) receptor alpha (extracellular domain), derived from the insect cell line Sf21.
• Specificity: NB100-65840 is specific for PDGF receptor alpha. The ND50 was determined as 0.2-0.6 ug/ml for blocking 10 ng/ml of PDGF-AA biological activity on a quiescent confluent cell culture of human WS1 cells. PDGF is a major serum mitogen that can exist as a homo- or heterodimeric protein consisting of disulphide-linked PDGF-A and PDGF-B chains. The PDGF-AA, PDGF-BB and PDGF-AB isoforms have been shown to bind to two distinct cellsurface PDGF receptors with different affinities. Whereas PDGF receptor alpha binds all three isoforms with high affinity, PDGF R beta binds PDGF-BB and AB but not PDGF-AA. Both PDGF receptor alpha and beta are members of the class III subfamily of receptor tyrosine kinases.

Background

Platelet derived growth factor receptor (PDGF Receptor) is a transmembrane glycoprotein of 170-185 kDa which undergoes homo or heterodimerization into complexes of alpha and beta subunits upon ligand binding, depending on the isoform of PDGF (PDGF AA, BB or AB) that binds. The phosphorylation of tyrosine residues in the now activated receptor can control multiple signaling events such as actin reorganization, transcription, cell growth, migration and differentiation, and also lead to activation of the Ras, Raf and ERK 1 and 2 pathway. This Ab blocks PDGF Receptor alpha mediated biological activity induced by PDGF-AA.
