Western Blot: Nucleostemin Antibody (JG37-82) [NBP2-76853] - Analysis of Nucleostemin on A549 cell lysates with Rabbit anti-Nucleostemin antibody at 1/500 dilution. Lysates/proteins at 10 ug/Lane. Predicted band size: 62 kDa Observed band size: 62 kDa Exposure time: 2 minutes; 8% SDS-PAGE gel. Proteins were transferred to a PVDF membrane and blocked with 5% NFDM/TBST for 1 hour at room temperature. The primary antibody at 1/500 dilution was used in 5% NFDM/TBST at room temperature for 2 hours. Goat Anti-Rabbit IgG - HRP Secondary Antibody at 1:300,000 dilution was used for 1 hour at room temperature.

Immunocytochemistry/Immunofluorescence: Nucleostemin Antibody (JG37-82) [NBP2-76853] - ICC staining Nucleostemin in HepG2 cells (green). The nuclear counter stain is DAPI (blue). Cells were fixed in paraformaldehyde, permeabilised with 0.25% Triton X100/PBS.

Immunohistochemistry-Paraffin: Nucleostemin Antibody (JG37-82) [NBP2-76853] - Analysis of paraffin-embedded human colon carcinoma tissue with Rabbit anti-Nucleostemin antibody washed with ddH2O and PBS, and then probed with the primary antibody at 1/200 dilution for 1 hour at room temperature. The detection was performed using an HRP conjugated compact polymer system. DAB was used as the chromogen. Tissues were counterstained with hematoxylin and mounted with DPX.

Flow Cytometry: Nucleostemin Antibody (JG37-82) [NBP2-76853] - Flow cytometric analysis of A549 cells with Nucleostemin antibody at 1/50 dilution (Fuchsia) compared with an unlabelled control (cells without incubation with primary antibody; Yellow). Alexa Fluor 488-conjugated goat anti-rabbit IgG was used as the secondary antibody.

Background

Nucleostemin is a protein found in the nucleoli of embryonic stem cells, adult CNS stem cells, primitive cells in the bone marrow and cancer cells. It is not in the differentiated cells of most adult tissues. It has been suggested to play a role in controlling the cell-cycle progression in stem cells and cancer cells.
