Background
Neurogenic locus notch homolog protein 4 (Notch4) is a member of the Notch family. The Notch4 protein has an extracellular domain that contains several epidermal growth factor-like (EGF) repeats and an intracellular domain that contains a multitude of domain types. The Notch4 signaling network is an evolutionarily conserved intracellular pathway that regulates physical cell to cell interactions. The Notch4 protein, once cleaved from the trans-Golgi network, is presented on the cell surface as a heterodimer. Notch4 regulates cell-fate determination as a receptor for membrane-bound ligands Jagged1, Jagged2 and Delta1. In Drosophila, once these ligands are activated via the release of the notch intracellular domain (NICD), Notch4 forms a transcriptional activator complex with RBPJ/RBPSUH which then activates the genes of the Enhancer of Split (E(spl)) locus. Notch4 also plays a role in vascular, renal and hepatic development, the implementation of differentiation, proliferation and apoptotic programs, and the regulation of branching morphogenesis in the development of the vascular system. Notch4 is highly expressed in heart tissue, with moderate to low expression seen in lung, placenta, liver, skeletal muscle, kidney, pancreas, spleen, lymph node, thymus, bone marrow and fetal liver. Notch4 was not seen expressed in adult brain or peripheral blood leukocytes. Polymorphisms in the Notch4 gene are strongly associated with susceptibility to schizophrenia by altering either the efficiency or alternate splicing of Notch4 transcripts (PMID: 22488909). I have ordered NB100-93551. What is the positive control for the antibody?
Here are some cell lines that have good expression of this protein: Protein Atlas. I would recommend using something with strong expression as your positive control. From the site, HeLa, MCF7, NTERA2, Hep-G2, etc. all have strong expression of the protein and therefore would be good controls. These are all very common cell lines and ones we use in testing frequently.
