Western Blot: NAPE-PLD Antibody [NB110-80070] - Expression and activity of fatty acid amide hydrolase (FAAH) and NAPE-PLD in DRGs and spinal cord after nerve injury. QRT-PCR and Western Blot analysis of NAPE-PLD mRNA and protein expression in DRGs 7 days after SNI and treatment with R-flurbiprofen 4.5 mg/kg twice daily or vehicle. For QRT-PCR and Western Blot analysis pooled samples of each 3 mice were used and two independent experiments were performed. Background

N-Acylphosphatidylethanolamine (NAPE)-hydrolyzing phospholipase D (NAPE-PLD) is a membrane-bound enzyme which releases the endocannabinoid anandamide and other bioactive N-acylethanolamines from their corresponding NAPEs in animal tissues. NAPE-PLD has been shown to be expressed by specific populations of neurons in the brain and targeted to axonal processes. Recent studies have suggested that NAEs generated by NAPE-PLD in axons may act as anterograde synaptic signaling molecules that regulate the activity of postsynaptic neurons. NAPE-PLD has also been shown to catalyze the hydrolysis of NAPE to generate OEA (oleoylethanolamide), a natural lipid amide that inhibits food intake in free-feeding rodents by prolonging latency to feed and postmeal interval.

  NAPE-PLD antibody used for WB application in experiments involving the evaluation of endocannabinoid system (ECS) in black truffle Tuber melanosporum. 15 ug/lane of truffle homogenates prepared from powdered truffles (T. melanosporum) were analyzed on 10% SDS-PAGE with NAPE-PLD antibody used at 1:500 dilution. Lysate of mouse brain was included as a positive control and the antibody detected the target band around 46kDa (Fig. 1b).
