Immunocytochemistry/ Immunofluorescence: Myelin PLP Antibody [NB100-1608] - Immunohistochemistry photomicrograph of PLP immunoreactivity (Green; 1:1000 dilution) in a cryostat section of an embryonic (e18) mouse brain. ...read more
Immunohistochemistry-Frozen: Myelin PLP Antibody [NB100-1608] - Human brain and spinal cord stained with Myelin PLP antibody (green) at 1:3000 dilution.
Chickens were immunized with a synthetic peptide/keyhole limpet hemocyanin (KLH) conjugate. The peptide corresponded to a region of Myelin PLP, but are shared between the human (NP_000524, PLP1 isoform 1 and PLP1 isoform 2 (NP_955772)] and mouse (P60202).

Quality assurance analysis was performed using immunohistochemistry (1:2000) using fluorescein-labeled goat anti-chicken IgY (1:500) as the secondary reagent. Western Blot reactivity reported in scientific literature (PMID: 24057669).
30 kDa. Disclaimer note: The observed molecular weight of the protein may vary from the listed predicted molecular weight due to post translational modifications, post translation cleavages, relative charges, and other experimental factors.

Purification Notes After repeated injections into the hens, immune eggs were collected, and the IgY fractions were purified from the yolks. These IgY fractions were then affinity-purified using a peptide column, the concentration of the eluate adjusted to 100 ug/ml, and the preparation was filter sterilized.
Storage Notes Store at 4C in the dark. Under these conditions, the antibodies should have a shelf life of at least 12 months (provided they remain sterile). Do not freeze these antibodies unless you want to store them for longer periods of time. Note, however, that each time an antibody preparation is frozen, about half of its binding activity is lost.

Human PLP (also known as lipophilin) is a 30 kDa protein (277 amino acids) found in myelin of the CNS and PNS. Expressed by oligodendrocytes and Schwann cells, PLP stabilizes myelin by preventing lipid bilayer fusion, and aids in compaction. Different mutations of the human PLP gene product result in two neurological disorders - Pelizaeus-Merzbacher disease and Spastic Paraplegia type 2.
