Chickens were immunized with a peptide / keyhole limpet hemocyanin (KLH) conjugate. The synthetic peptide corresponded to a region shared between the mouse (P60202, NCBI) and human gene products [PLP1 isoform 1 (NP_000524, NCBI), PLP1 isoform 2 (NP_955772, NCBI)]. The sequence can be found between amino acid 26-75. After repeated injections into the hens, immune eggs were collected, and the IgY fractions were purified from the yolks. These IgY fractions were then affinity-purified using a peptide column, the concentration of the eluate adjusted to 100 ug/ml, and the preparation was filter-sterilized.

Human PLP (also known as lipophilin) is a 30 kDa protein (277 amino acids) found in myelin of the CNS and PNS. Expressed by oligodendrocytes and Schwann cells, PLP stabilizes myelin by preventing lipid bilayer fusion, and aids in compaction. Different mutations of the human PLP gene product result in two neurological disorders - Pelizaeus-Merzbacher disease and Spastic Paraplegia type 2.
