Specificity
NB100-65541 recognizes a monomorphic determinant of the rat I-A antigen present on B lymphocytes, dendritic cells, some macrophages and certain epithelial cells. This antibody cross reacts with certain mouse strains of MHC haplotypes k and s. This antibody does not react with C57BL6 or C57BL6/j mice because they are haplotype b. Analysis of recombinant mouse strains showed that the determinants mapped to the I-A region. The clone does not react with Rat BDIX strain. Please be aware of the possible size variance seen in western blot.

For IHC-Paraffin: This product requires antigen retrieval using heat treatment prior to staining of paraffin sections.Sodium citrate buffer pH 6.0 is recommended for this purpose. PLP fixation is recommended for optimal results.

Background

MHC Class II antigens are heterodimers consisting of one alpha chain(31-34kD) and one beta chain (26-29kD). The family of monoclonal antibodies (ER-TR 3, ER-TR 2, ER-TR 1) detect MHC class II antigens encoded by the murine Ia region of the H-2 complex, corresponding to the human HLA-DR region. MHC Class II antigens are a valuable tool for studying T helper cell interaction with class II positive antigen presenting cells (dendritic cells, B cells, macrophages) and offer new possibilities for studying the development of T helper cells since these antibodies also stain stromal cells in the thymus. MHC Class II antigens are also inducible on a number of other cells (endothelium and epithelial cells) by interferon gamma.

  1. Does this antibody recognize MHCII from the C57BL6 strain?
    • I heard back from our lab to your question on haplotype vs reactivity of MHC Class II Antibody (OX-6) [NB100-65541]. He told me that we would not expect this antibody to recognize C57BL6 or C57BL6/j since they are haplotype b.
