Immunocytochemistry /Immunofluorescence: MHC Class II Antibody (OX-6) - Chimeric - BSA Free [NBP3-43539] - Immunofluorescence analysis of paraformaldehyde fixed rat (Rattus norvegicus) splenocytes on Shi-fix coverslips, permeabilized with 0.15% Triton stained with NBP3-43539 at 10 ug/ml for 1h followed by Alexa Fluor488 secondary antibody (1 ug/ml), showing membrane and some cytoplasmic staining of a subset of cells. The nuclear stain is DAPI (blue). Panels show from left-right, top-bottom NBP3-43539, DAPI, merged channels and an isotype control. The isotype control was stained with an anti-Fluorescein antibody followed by Alexa Fluor488 secondary antibody.

Flow Cytometry: MHC Class II Antibody (OX-6) - Chimeric - BSA Free [NBP3-43539] - Staining of rat (Rattus norvegicus) leukocytes with either an isotype control (A) or NBP3-43539(B) at a concentration of 1 ug/ml for 30 mins at RT. After washing, bound antibody was detected using an Alexa Fluor 488 conjugated donkey anti-rabbit antibody and cells analysed on a FACSCanto flow-cytometer.

Western Blot: MHC Class II Antibody (OX-6) - Chimeric - BSA Free [NBP3-43539] - Rat spleen (A), rat thymus (B) and rat lymph node (C) lysate samples (35ug protein in RIPA buffer) were resolved on a 10% SDS PAGE gel and blots probed with NBP3-43539 at 1 ug/ml before detection using an anti-rabbit secondary antibody. A primary incubation of 1h was used and protein was detected by chemiluminescence. The expected running size for CD74 is ~24.5-31.6kDa, depending on the isoform and post-translational modification. NBP3-43539 successfully detected CD74 in rat spleen, thymus and lymph node lysates.

Background

MHC Class II antigens are heterodimers consisting of one alpha chain(31-34kD) and one beta chain (26-29kD). The family of monoclonal antibodies (ER-TR 3, ER-TR 2, ER-TR 1) detect MHC class II antigens encoded by the murine Ia region of the H-2 complex, corresponding to the human HLA-DR region. MHC Class II antigens are a valuable tool for studying T helper cell interaction with class II positive antigen presenting cells (dendritic cells, B cells, macrophages) and offer new possibilities for studying the development of T helper cells since these antibodies also stain stromal cells in the thymus. MHC Class II antigens are also inducible on a number of other cells (endothelium and epithelial cells) by interferon gamma.
