Western Blot: FXR/NR1H4 Antibody [NB400-153] - Total protein from human A549, U2OS cell and mouse 3T3 cells was separated on a 7.5% gel by SDS-PAGE, transferred to PVDF membrane and blocked in 5% non-fat milk in TBST. The membrane was probed with 2.0 ug/ml anti-FXR (NB400-153) in block buffer and detected with an anti-rabbit HRP secondary antibody using NovaLume chemiluminescence detection reagent (NPB2-61915).
Immunocytochemistry/Immunofluorescence: FXR/NR1H4 Antibody [NB400-153] - A549 cells were fixed for 10 minutes using 10% formalin and then permeabilized for 5 minutes using 1X PBS + 0.5% Triton-X100. The cells were incubated with anti-FXR/NR1H4 at 2 ug/ml overnight at 4C and detected with an anti-rabbit Dylight 488 (Green) at a 1:500 dilution. Alpha tubulin (DM1A) NB100-690 was used as a co-stain at a 1:1000 dilution and detected with an anti-mouse Dylight 550 (Red) at a 1:500 dilution. Nuclei were counterstained with DAPI (Blue). Cells were imaged using a 40X objective.

Dual RNAscope ISH-IHC: ABCA1 Antibody [NB400-105] - Formalin-fixed paraffin-embedded tissue sections of human prostate cancer were probed for ABCA1 mRNA (ACD RNAScope Probe, catalog # 432291; Fast Red chromogen, ACD catalog # 322360). Adjacent tissue section was processed for immunohistochemistry using rabbit polyclonal (Novus Biologicals catalog # NB400-105) at 1.5ug/mL with overnight incubation at 4 degrees Celsius followed by incubation with anti-rabbit IgG VisUCyte HRP Polymer Antibody (Catalog # VC003) and DAB chromogen (yellow-brown). Tissue was counterstained with hematoxylin (blue). Specific staining was localized to glandular cells.
Western Blot: ABCA1 Antibody [NB400-105] - Analysis of ABCA1 in total cell lysates of RAW264.9 cells treated with vehicle (-) or 9-cisretinoic acid and 22Rhydroxycholesterol (+). Samples used for this testing were 40 ug of total cell post-nuclear lysate from each group.

The cells were incubated with anti-ABCA1 at 5.0ug/ml overnight at 4C and detected with an anti-rabbit Dylight 488 (Green) at a 1:500 dilution. Alpha tubulin (DM1A) NB100-690 was used as a co-stain at a 1:1000 dilution and detected with an anti-mouse Dylight 550 (Red) at a 1:500 dilution. Nuclei were counterstained with DAPI (Blue). Western Blot: PPAR alpha/NR1C1 Antibody [NB600-636] - Affinity Purified Detection a 52 kDa band corresponding to PPAR alpha present in a 3T3 whole cell lysate. Approximately 20 ug of lysate was loaded per lane for SDS-PAGE. Detection occurred after using a 1:500 (lane 1) or 1:1000 (lane 2) dilution of antibody followed by 1:2000 dilution of HRP Goat-a-Rabbit IgG for visualization. Storage Conditions: Store vial at -20 C prior to opening. Dilute only prior to immediate use. For extended storage aliquot contents and freeze at -20 C or below. Avoid cycles of freezing and thawing. Expiration date is one (1) year from date of opening.

Immunohistochemistry-Paraffin: PPAR alpha/NR1C1 Antibody [NB600-636] - Rat brain sections, highlighting cytoplasmic staining in ependymal cells and neurons in frontal cortex. Bottom image shows subventricular zone (svz) of lateral ventricle (exit point of progenitor olfactory neurons); top image shows frontal cortex in the same section. Cytoplasmic staining is also observed in the corpus callosum (bottom image) and in dendritic fields of the cortex. Formalin/PFA-fixed paraffin-embedded sections of rat brain tissue were incubated with the primary antibody at 1:200 for 1 hour. Antigen retrieval was performed by heat induction in citrate buffer pH 6.0.

Immunohistochemistry: Insulin Antibody (E2-E3 (same as INS04)) [NBP2-34260] - Immunohistochemistry of pancreas shows relatively higher reactivity for insulin in PrP-/- relative to Tg40 PrP sections (panels 1 & 3). Iron overloading decreases insulin reactivity in Tg40 PrP but not in PrP-/- samples (panels 1 vs. 2 & 3 vs. 4). Reaction for glucagon is higher in iron overloaded Tg40 PrP relative to control samples (panels 5 & 6). The difference is minimal in PrP-/- samples (panels 7 & 8). Immunohistochemistry-Paraffin: VDR/NR1I1/Vitamin D Receptor Antibody (JA11-16) [NBP2-66778] - Analysis of paraffin-embedded rat colon tissue using anti-Vitamin D Receptor antibody. The section was pre-treated using heat mediated antigen retrieval with sodium citrate buffer (pH 6.0) for 20 minutes. The tissues were blocked in 5% BSA for 30 minutes at room temperature, washed with ddH2O and PBS, and then probed with the antibody at 1/100 dilution, for 30 minutes at room temperature and detected using an HRP conjugated compact polymer system. DAB was used as the chromogen. Counter stained with hematoxylin and mounted with DPX.
