Background

This gene encodes a member of the lysyl oxidase gene family. The prototypic member of the family is essential to the biogenesis of connective tissue, encoding an extracellular copper-dependent amine oxidase that catalyses the first step in the formation of crosslinks in collagens and elastin. A highly conserved amino acid sequence at the C-terminus end appears to be sufficient for amine oxidase activity, suggesting that each family member may retain this function. The N-terminus is poorly conserved and may impart additional roles in developmental regulation, senescence, tumor suppression, cell growth control, and chemotaxis to each member of the family.

Immunohistochemistry-Paraffin: Lysyl Oxidase Homolog 3/LOXL3 Antibody [NBP2-75964] - Lysyl Oxidase Homolog 3/LOXL3 [NBP2-75964] - IHC analysis of a formalin fixed paraffin embedded tissue section of human small intestine using 1:200 dilution of LOXL3 antibody (NBP2-75964). The signal was developed using HRP-DAB method which followed counterstaining of the cells with hematoxylin. The antibody generated cytoplasm and membrane staining of glandular cells.
Western Blot: Lysyl Oxidase Homolog 3/LOXL3 Antibody [NBP2-75964] - Total protein from human A431, HepG2, mouse Neuro2A and rat PC12 cell lines was separated on a 4-20% gel by SDS-PAGE, transferred to PVDF membrane and blocked in 5% non-fat milk in TBST. The membrane was probed with 2.0 ug/ml anti-LOXL3 in block buffer and detected with an anti-rabbit HRP secondary antibody using West Pico PLUS chemiluminescence detection reagent.
Immunocytochemistry/Immunofluorescence: Lysyl Oxidase Homolog 3/LOXL3 Antibody [NBP2-75964] - A431 cells were fixed for 10 minutes using 10% formalin and then permeabilized for 5 minutes using 1X PBS + 0.5% Triton-X100. The cells were incubated with anti-LOXL3 at 10 ug/ml overnight at 4C and detected with an anti-rabbit Dylight 488 (Green) at a 1:500 dilution. Nuclei were counterstained with DAPI (Blue). Cells were imaged using a 40X objective.

Western blot shows lysates of human uterus tissue and human heart tissue. PVDF membrane was probed with 0.1 µg/mL of Goat Anti-Human Decorin Antigen Affinity-purified Polyclonal Antibody (Catalog # AF143) followed by HRP-conjugated Anti-Goat IgG Secondary Antibody (Catalog # HAF017). A specific band was detected for Decorin at approximately 100 kDa (as indicated). This experiment was conducted under reducing conditions and using Immunoblot Buffer Group 1.
Simple Western lane view shows lysates of human uterus and human heart tissue, loaded at 0.2 mg/mL. A specific band was detected for Decorin at approximately 140-149 kDa (as indicated) using 5 µg/mL of Goat Anti-Human Decorin Antigen Affinity-purified Polyclonal Antibody (Catalog # AF143). This experiment was conducted under reducing conditions and using the 12-230 kDa separation system.        
Decorin was detected in immersion fixed paraffin-embedded sections of human breast using Goat Anti-Human Decorin Antigen Affinity-purified Polyclonal Antibody (Catalog # AF143) at 15 µg/mL overnight at 4 °C. Tissue was stained using the Anti-Goat HRP-DAB Cell & Tissue Staining Kit (brown; Catalog # CTS008) and counterstained with hematoxylin (blue). Cells were either treated with 3uM Monensin for 3 hours to block the secretion of Complement C3 (B) or grown in normal media (A). Cells were fixed with 4% PFA and then permeablized with 0.1% saponin. Cells were incubated in an antibody dilution of 2 ug/mL for 30 minutes at room temperature, followed by mouse F(ab)2 IgG (H+L) PE-conjugated secondary antibody (F0102B, R&D Systems).
Immunocytochemistry/Immunofluorescence: Complement C3 Antibody (11H9) [NB200-540] - C3 protein fragments deposited on kidney cells of MPL-lpr mouse. Staining with antibody 11H9. Glomerular staining pattern. Fixation in 4% paraformaldehyde in PBS pH 7.4. Vibratome sections of 4 um. Pretreated with 3% hydrogen peroxide for 20 min to quench endogenous peroxidases. Microwaved in antigen unmasking solution for 2-5 minutes as antigen retrieval.
Immunohistochemistry-Paraffin: Complement C3 Antibody (11H9) [NB200-540] - Complement C3 protein in a FFPE tissue section of mouse liver using 1:100 dilution of Complement C3 antibody (clone 11H9) NB200-540. Weak but distinct membrane-cytoplasmic immunopositivity was observed in hepatocytes and few cells developed punctate membrane staining.
