Background

SUV39H1 (suppressor of variegation 3-9 homolog 1) is a histone methyltransferase that contains a chromodomain and a C-terminal SET domain. It specifically trimethylates histone H3 at lys 9. Interaction of SUV39H1 with RB1 results in recruitment to histone H3. Trimethylation of H3 stimulates the binding of heterochromatin proteins ((CBX1, CBX3 and/or CBX5) which leads to epigenetic transcriptional repression in heterochromatin regions. Recent research has also shown that this protein interacts with oncogenes involved in AML. Treatment of AML cells with chaetocin caused induction of apotosis via direct and indirect modulation of SUV39H1 activity (PMID:21979880). This protein plays a critical role in heterochromatin organization, chromosome segregation, and mitotic progression.
