Western Blot: Islet-1 Antibody (SC05-64) [NBP2-67576] - Analysis of Islet-1 on MCF-7 cell lysates with Rabbit anti-Islet 1 antibody at 1/500 dilution. Lysates/proteins at 10 ug/Lane. Predicted band size: 39 kDa Observed band size: 42 kDa Exposure time: 2 minutes; 10% SDS-PAGE gel. Proteins were transferred to a PVDF membrane and blocked with 5% NFDM/TBST for 1 hour at room temperature. The primary antibody at 1/500 dilution was used in 5% NFDM/TBST at room temperature for 2 hours. Goat Anti-Rabbit IgG - HRP Secondary Antibody at 1:300,000 dilution was used for 1 hour at room temperature.

Immunocytochemistry/Immunofluorescence: Islet-1 Antibody (SC05-64) [NBP2-67576] - Staining Islet 1 in A549 cells (green). The nuclear counter stain is DAPI (blue). Cells were fixed in paraformaldehyde, permeabilised with 0.25% Triton X100/PBS.

Background

Islet 1 encodes a member of the LIM/homeodomain family of transcription factors. The encoded protein binds to the enhancer region of the insulin gene, among others, and may play an important role in regulating insulin gene expression. The encoded protein is central to the development of pancreatic cell lineages and may also be required for motor neuron generation. Mutations in this gene have been associated with maturity-onset diabetes of the young.
