IRF8 Antibody [CoraFluor™ 1]

CoraFluor™ 1, amine reactive (Catalog:7920) and CoraFluor™ 2, amine reactive (Catalog # 7950) are terbium-based probes that have been developed for use as TR-FRET donors. They emit wavelengths compatible with commonly used fluorescent acceptor dyes such as BODIPY® (or BDY) and Janelia Fluor® dyes, FITC (Catalog # 5440), TMR and Cyanine 5 (Catalog # 5436). CoraFluor™ fluorescence is brighter and more stable in biological media than existing TR-FRET donors, leading to enhanced sensitivity and improved data generation. CoraFluor™ 1 exhibits excitation upon exposure to a 337 nm UV laser.

• Description: CoraFluor(TM) 1 is a high performance terbium-based TR-FRET (Time-Resolved Fluorescence Resonance Energy Transfer) or TRF (Time-Resolved Fluorescence) donor for high throughput assay development. CoraFluor(TM) 1 absorbs UV light at approximately 340 nm, and emits at approximately 490 nm, 545 nm, 585 nm and 620 nm. It is compatible with common acceptor dyes that absorb at the emission wavelengths of CoraFluor(TM) 1. CoraFluor(TM) 1 can be used for the development of robust and scalable TR-FRET binding assays such as target engagement, ternary complex, protein-protein interaction and protein quantification assays.

Background

Interferon regulatory factor 8 (IRF8) / Interferon consensus sequence binding protein (ICSBP) is a transcription factor of the interferon (IFN) regulatory factor (IRF) family. Proteins of this family are composed of a conserved DNA binding domain in the N terminal region and a divergent C terminal region that serves as the regulatory domain. The IRF family proteins bind to the IFN stimulated response element (ISRE) and regulate expression of genes stimulated by type I IFNs, namely IFN alpha and IFN beta. IRF family proteins also control expression of IFN alpha and IFN beta regulated genes that are induced by viral infection. IRF8 specifically binds to the upstream regulatory region of type I IFN and IFN inducible MHC class I genes (the interferon consensus sequence (ICS)). Unlike IRF3, IRF8 appears to act as a negative regulator of IFN induced genes in most cases, but IRF8 mediates activation of NF kappaB by the toll like receptor 9 (TLR9) after stimulation by unmethylated CpG DNA in dendritic cells. Finally, it has been shown that IRF8 decreases bcl-2 expression and thus may play a role in chronic myelogenous leukemia.
