We are interested in your antibody IL17RD Antibody (NBP1-32307). However, we have a question regarding it. There are two WB images on your website, where you show the detection of IL17RD. There are two WB images on your website, where you show the detection of IL17RD. On the first image you point to an approximately 80kDa band in HEK293T cells. Is this a transfected protein or the endogenous one? If this is the endogeous IL17RD, why is it 80kDa, and not approximately 120kDa, as it is expected from a glycosylated transmembrane IL17RD? Or is this the cytosolic isoform 4 of IL17RD? Could you clarify this for us?
- In regards to your inquiry the HEK293T cells are transfected with the protein as an overexpression lysate. This product however is also tested on endogenous samples of 3T3 and Mouse brain. In those samples a band of similar size was also observed. Despite the glycosylation of this protein we detected it at approx. 79 kDa in our samples. The antibody should not be specific for just isoform 4. Since it is picking up at the lower molecular weight it may be detecting the cytosolic form with more prevalence than the membrane bound protein since whole cell preps were used and not just membrane preps.
