Capture ELISA: this antibody can be used as a capture antibody in a rat IL-1a ELISA with a biotinylated detection antibody. It is recommended to use plates coated with 100 mL/well of the capture antibody at a concentration of 1 mg/mL. Immunoblotting: a working concentration of 1-2 ug/mL is recommended using 50 ng/lane of rat IL-1a under either reducing or non-reducing conditions.
Cross-reacts with Rat.Not yet tested in other species.

Interleukins (ILs) are a large group of cytokines that are produced mainly by leukocytes, although some are produced by certain phagocytes and auxiliary cells. Each IL acts on a specific, limited group of cells through a receptor specific for that IL. Interleukin 1 (IL1), originally known as lymphocyte activating factor (LAF), activates T cells and lymphocytes, which then proliferate and secrete interleukin 2. IL1 is primarily released from stimulated macrophages and monocytes, but also is released from several other cell types and is thought to play a key role in inflammatory and immune responses. The two closely related agents, interleukin 1 alpha (IL1 alpha) and interleukin 1 beta (IL1 beta) bind to the same cell surface receptor, elicit nearly identical biological responses and share 25% homology in their amino acid sequence. I read about one of your other IL1 beta antibodies that primarily detects the mature 17 kDa protein, but also to some extent the pro-protein which is cleaved to the 17kDa product by the heating applied in the preparation of the samples for WB. Is this also the case for this antibody (NB120-10747) or does it only detect the "biologically" cleaved protein?
- Excessive heating or prolonged heating of your samples can cause peptide bond cleavage and protein degradation. It is impossible to differentiate between the biologically cleaved and heat induced peptide bond cleavage. For detecting proforms of protein, following addition of SDS sample buffer, the sample should be heated to 85C for 3 minutes. The standard western blot protocol calls for boiling (95C) for 10 minutes however in this case decreasing the time and temperature will help detect the proform of the protein.
