The signal was developed using HRP-conjugated anti-rat secondary with DAB reagent which followed counterstaining of nuclei using hematoxylin. This antibody specifically labelled the endothelial cells in blood vessels located primarily in the sub-mucosa, and of that of the mucosa muscularis and the mucosal lacteal.

Western blot shows lysates of MO7e human megakaryocytic leukemic cell line, P815 mouse mastocytoma cell line, and MC/9‑2 mouse mast cell line. PVDF membrane was probed with 0.1 µg/mL of Goat Anti-Human/Mouse CD117/c-kit Antigen Affinity-purified Polyclonal Antibody (Catalog # AF1356) followed by HRP-conjugated Anti-Goat IgG Secondary Antibody (Catalog # HAF017). Specific bands were detected for CD117/c-kit at approximately 135, 150 kDa (as indicated). This experiment was conducted under reducing conditions and using Immunoblot Buffer Group 1.
Simple Western lane view shows lysates of MO7e human megakaryocytic leukemic cell line, P815 mouse mastocytoma cell line and MC/9-2 mouse mast cell line, loaded at 0.2 mg/mL. A specific band was detected for CD117/c-kit at approximately 150-165 kDa (as indicated) using 5 µg/mL of Goat Anti-Human/Mouse CD117/c-kit Antigen Affinity-purified Polyclonal Antibody (Catalog # AF1356) followed by 1:50 dilution of HRP-conjugated Anti-Goat IgG Secondary Antibody (Catalog # HAF109). This experiment was conducted under reducing conditions and using the 12-230 kDa separation system.        
CD117/c-kit was detected in immersion fixed frozen sections of mouse embryo using Goat Anti-Human/Mouse CD117/c-kit Antigen Affinity-purified Polyclonal Antibody (Catalog # AF1356) at 15 µg/mL overnight at 4 °C. Tissue was stained using the Anti-Goat HRP-DAB Cell & Tissue Staining Kit (brown; Catalog # CTS008) and counterstained with hematoxylin (blue). 1 ug/lane of Recombinant Human BMP-4 was resolved with SDS-PAGE under reducing (R) and non-reducing (NR) conditions and visualized by silver staining, showing major bands at 22-25 kDa and 37-41 kDa, respectively. Multiple bands in gel are due to variable glycosylation. 
Recombinant Human BMP-4 (Catalog #314-BP/CF) induces alkaline phosphatase production in the ATDC5 mouse chondrogenic cell line. The ED50 for this effect is 2.5-15 ng/mL.

Western Blot: NOD2 Antibody (2D9) [NB100-524] - Total protein from human THP1 and US-OS cells and mouse Raw264.7 cells was separated on a 7.5% gel by SDS-PAGE, transferred to PVDF membrane and blocked in 5% non-fat milk in TBST. The membrane was probed with 2.0 ug/ml anti-NOD2 in 1% non-fat milk in TBST and detected with an anti-mouse HRP secondary antibody using chemiluminescence.
Immunocytochemistry/Immunofluorescence: NOD2 Antibody (2D9) [NB100-524] - A431 cells were fixed for 10 minutes using 10% formalin and then permeabilized for 5 minutes using 1X TBS + 0.5% Triton X-100. The cells were incubated with anti-NOD2 (2D9) NB100-524 at a 1:200 dilution overnight at 4C and detected with an anti-mouse DyLight 488 (Green) at a 1:500 dilution. Actin was counterstained with Phalloidin 568 (Red) at a 1:200 dilution. Nuclei were counterstained with DAPI (Blue). Cells were imaged using a 40X objective.
