Flow Cytometry: Histamine H3R Antibody (27H8) [NBP3-26219] - Overlay Peak curve showing SH-SY5Y cells stained with Histamine H3R Antibody (27H8) (red line) at 1:50. The cells were fixed in 4% formaldehyde and permeated by 0.2% Triton X-100. Then 10% normal goat serum to block non-specific protein-protein interactions followed by the antibody (1ug/1*10^6 cells) for 45min at 4C. The secondary antibody used was FITC-conjugated Goat Anti-rabbit IgG (H+L) at 1:200 dilution for 35min at 4C. Control antibody (green line) was rabbit IgG (1ug/1*10^6 cells) used under the same conditions. Acquisition of >10,000 events was performed.

Background

H3 Histamine Receptor regulates the release of the neurotransmitters norepinephrine and somatostatin. In histamine-containing neurons of the brain, the H3 receptor acts as a presynaptic autoreceptor, while in nonhistamine-containing neurons in both the central and peripheral nervous systems, it acts as a presynaptic heteroreceptor. Multiple isoforms of H3 receptors are produced by alternative splicing. Histamine H3 receptor has been reported primarily in brain. ESTs have been isolated from brain (normal and cancer), breast cancer, colon cancer, and retinoblastoma libraries.
