Flow Cytometry: hHR23b Antibody (SAIC-28A-29) - Azide and BSA Free [NBP3-20096] - Paraformaldehyde fixed HeLa cells permeabilized with 0.5% Triton were stained with anti-unknown specificity antibody (isotype control, black line) or the rabbit IgG version of SAIC-28A-29 (NBP3-20096, blue line) at a dilution of 1:100 for 1h at RT. After washing, the bound antibody was detected using a goat anti-rabbit IgG AlexaFluor(R) 488 antibody at a dilution of 1:1000 and cells analyzed using a FACSCanto flow-cytometer.

Actin filaments were stained with phalloidin (red) and the nuclear stain is DAPI (blue). Panels show from left-right, top-bottom NBP3-20096, DAPI, merged channels and an isotype control. The isotype control was an unknown specificity antibody followed by staining with Alexa Fluor(R) 488 secondary antibody.

Background

hHR23b is encoded by this gene is one of two human homologs of Saccharomyces cerevisiae Rad23, a protein involved in the nucleotide excision repair (NER). This protein was found to be a component of the protein complex that specifically complements the NER defect of xeroderma pigmentosum group C (XP-c) cell extracts in vitro. This protein was also shown to interact with, and elevate the nucleotide excision activity of 3-methyladenine-DNA glycosylase (MPG), which suggested a role in DNA damage recognition in base excision repair. This protein contains an N-terminal ubiquitin-like domain, which was reported to interact with 26S proteasome, and thus this protein may be involved in the ubiquitin mediated proteolytic pathway in cells.
