ChIP usage was reported in scientific literature (PMID: 17116691; 2007) WB: Detects an approx. 97 kDa protein representing GR from rat liver extract. ICC: Staining of GR in HeLa cells results in cytoplasmic staining in the absence of ligand, and nuclear staining after hormone administration.

Background

Steroid receptors are ligand-dependent, intracellular proteins that stimulate transcription of specific genes by binding to specific DNA sequences following activation by the appropriate hormone. Glucocorticoids are a family of steroids necessary for the regulation of energy metabolism and the immune and inflammatory responses. These compounds exert their effect through their interaction with the glucocoticoid receptor (GR) and that complex's subsequent association with DNA. All normal mammalian tissues examined to date have been shown to contain glucocorticoid receptor.

Electrophoresis was carried out using 35ug of whole cell lysate with a 4-12% Bis-Tris NuPage gel and NuPage MES running buffer. The blot was cut between 98 and 62 kDa. 0.5ug/mL glucocorticoid receptor antibody (NB300-610) was used in 5% milk TBST and incubated overnight at 4-degrees. The secondary antibody (GE healthcare) was used at 1:20 000. The blot shows the glucocorticoid receptor alpha variants A, B and C.
