Western Blot: ER beta/NR3A2 Antibody [NBP2-81132] - Total protein from human Adrenal gland, testis and tonsil was separated on a 7.5% gel by SDS-PAGE, transferred to PVDF membrane and blocked in 5% non-fat milk in TBST. The membrane was probed with 2.0 ug/ml anti-ER Beta in blocking buffer and detected with an anti-rabbit HRP secondary antibody using West Pico PLUS chemiluminescence detection reagent.

Immunohistochemistry-Paraffin: ER beta/NR3A2 Antibody [NBP2-81132] - ER beta / NR3A2 was detected in paraffin-embedded sections of human breast carcinoma tissue using 1:200 Rabbit Anti-Human ER beta / NR3A2 Antigen Affinity-purified Polyclonal Antibody (Catalog # NBP2-81132) overnight at 4 degrees C. Before incubation with the primary antibody tissue was subjected to heat-induced epitope retrieval using sodium citrate buffer (pH 6.0). Tissue was staining with DAB and counterstained with hematoxylin.

Background

Estrogen receptor beta (ER beta), a NR3 Steroid Receptor, has been shown to have physiological functions in the central nervous system, cardiovascular system, immune system, urogenital tract, gastrointestinal tract, and in bone, kidney, and lung. Mice lacking the gene for ER beta show defective ovary development, reduced fertility, abnormal bladder and prostate development, and impaired vascular system functions that lead to hypertension in aging. ER beta is phosphorylated upon ligand activation and binds to target DNA as a homodimer. At least eight alternatively spliced isoforms of ER beta have been documented, many of which show tissue-specific expression patterns. A novel and functionally distinct isoform of human ER beta (ER beta548), which includes an extended amino-terminal domain has been identified recently by Wilkinson et al. 2002. ER beta has been suggested to affect carcinogenesis in smooth muscle, urogenital tract, breast, and colonic tissues.
